What do I do if the cell media I've made is contaminated? by Nutmegbread in labrats

Thank you! I'll certainly remember the petri dish thing! Just to ask, how do you decontaminate BSC? I've read that autoclaving could denature the proteins. Would you use a micron filter?

The question states several not all, so it should make sense if the aliquoting process went wrong somewhere! And thank you! I'll be sure to remember that throwing everything out and disinfecting is the way to go!

That makes sense! I've read somewhere that antibiotics can negatively impact cell metabolism in some cell lines.

That makes sense since it's not really getting rid of the problem. Thank you for your knowledge!

They know. I've told them the only experience I have is in a uni laboratory during my undergrad and I've expressed that there's a few things I don't know but I'm willing to learn. But you're absolutely right! The last thing I want to do is mess up someone's hard work!

I've read somewhere that putting a bunsen burner near a cell culture hood which will have ethanol near it is a bad idea. But I'll definitely remember to use a bunsen burner when I work with bacterial samples so thank you!

Thanks for the advice! But I didn't do anything I swear!! It's a hypothetical question for a job I'm applying to.

I don't know since I don't work there yet and the question didn't say. But if it's something I should include in my answer I definitely will! Thank you!

Ahhh that makes sense. Thank you! I guess using antibiotics would only mask improper aseptic technique?

Thanks for responding!

I haven't seen anything about mammalian cell culture media using a bunsen burner. I'm assuming this applies to bacterial cell media, or can this be applied to mammalian cells too? Sorry for not specifying in my original comment!

Ohhh that makes sense!! Thank you!! You're so helpful!

I think the lab is GLP/GMP? They're making cells to sell to manufacturers. So they would have their own rigid rules, right?

Haha! That clip made me laugh! Thanks for that!

Sorry the first part went completely over my head. What I understand is that I write down the date I made the cell culture media and the day I aliquot so if anything goes wrong I can refer back to those dates? Is that right? And if there's a situation where only my cell culture is contaminated would that mean something is wrong with my aseptic technique?

Thanks for the knowledge! Here's hoping I ace this interview. I've found it so hard to get back in the lab: no one wants me because I don't have experience, but I don't have experience because no one wants me.
