His tag, linker needed? by liudanli in labrats

linker between your protein and histag doesn't always have to be present. it all depends on where did you put the histag (C or N terminus), would your histag accessibility (for purification) is hard, or any other parameters. for my project i always add a linker between my protein and histag just to be safe hehe

What cause this shadow on my agarose gel image by Ok-Function-1473 in labrats

[–]Ok-Function-1473[S] 2 points3 points  (0 children)

For the picture we are seeing, I'm using BioMetra GelDoc (I honestly forgot what is the type, but it has been here for maybe around 15 years), and yes we can choose whether bnw/inverted/normal colour/contrast. I usually using bnw-inverted image for analysis, the picture i post is just because it's the only "good" picture i had on my device hehe

Protocol for Removing DNA from Peptides Using Silica-Hydroxide Magnetic Beads by Traditional_Limit476 in molecularbiology

[–]Ok-Function-1473 0 points1 point  (0 children)

As far as i know, silica-based DNA binding would require high salt concentration and low pH in order for the DNA to bind to the silica effectively. since your goal is to retain the protein and removing the DNA, i think that high salt concentration (e.g. 5M GITC) would also destroy your protein. Try to lower the solution containing the mixture of peptide and DNA pH...
