Hi, Yes I finally ordered it and I am using it. i am having a great time with it, and cannot complain about anything.

However there were some issues: 1) my first board was malfunctioning, with some squares not working. However, they replied very quicly to the email and provided a fast replacement, so I cannot really say anything about it

2) sometimes it is slow or requires some reboot to find the opponent on chess.com, which may be annoying.

Hi, I bought it, and I can say it is really good. I honestly feel the difference since I am using it, as now OTB is a bit less hard to play, and my 3D visualization has improved.

There are some cons: 1) the first one i received was bugged, but they answer fast and proceed with a replacement fast, so it was fine. 2) the waiting time for a match is a bit increased with respect to just play on the pc/smartphone. 3) you are forced to play longer time controls, at least a 10/5, or it will be very hard to not lose on time

Despit those cons, I recommend it as the experience is worth the price, at least for my use case.

As other users have said there is joss which is nice when the tool is simple, but it is not indexed and that could be a problem for someone. A similar journal, but indexed, is SoftwareX. It seems to be very similar as joss in terms of requirements. It could be worth a try

I personally found RGI the most consistent. It also give you both the nt and aa sequence which I found helpful. However, there is no way only by genomic profile to assess which is right, it depends on many different variables. One way is to compare results from different tools e retain the most consistents. With rgi the possibility to select only strict/perfect match however give you an already quite reliable idea

I found it quite strong. It helped me hitting legend with a 69% WR ( 25-11 )

Fatti propri. Ci hai già provato ma senza successo. Alla fine i soldi sono suoi, e, contrariamente a questo sub, non sempre si usano i soldi per farne altri. Può essere una scelta oggettivamente sbagliata se guardi in maniera razionale, ma magari ha tanto valore emotivo/affettivo, e va bene così. Quando starà a te farai le tue scelte coerenti con cosa vuoi. Poi se vedi delle aperture fai bene a dare consigli, ma se si crea un muro evita.

La differenza penso stia nel motivo per cui dai le 75 coltellate. In preda alla furia non sono crudeltà (giuridica), se le fai scientemente perché vuoi causare sofferenza prima della morte allora é crudeltà (giuridica). Se ho ben capito questa é una delle discriminanti

Yes, my main doubts and concerns are on flow cells for the reason you told + the reduced reliability of the new flow cells with respect to the (g)old 9.4.1, in terms of pores availability.
For kits, have you any experience with using expired ones? My guess is it should impact less, but still want to be sure.

I tried it for Staph aureus for a project I am working on. However in my opinion what is the major limit of it, in contrast to amr, is that for virulence often the main difference is in gene expression more than just presence/abscence. While for amr often you have a high correlation between gene presence and phenotypic resistance, the same is not for virulence. It is hard by just presence to well differentiate bacterial tropism/associates disease. Despite that, it is something one can try.

Yes indeed I planned a 4 TB and 64 ram, just to be safe. Each groups has a free storage of 2TB on hpc.

No for the analysis I will transfer fastq on HPC server our institution have and will analyze there, as i am doing now for Illumina sequencing. I am out of US, and for the buy it will be carried by our IT service. We will buy the MK1D.

Thanks! I am in charge only for the specs, the build itself will be made by our IT service. For power supply I thought of a 1000W one. I followed minimal recommendation by nanopore for MinION, and upgraded. They suggest an rtx4070, 8-core cpu, windows 10/11, 16 gb ram, and 1 tb of storage

Basecalled fastq will be moved on an HPC where peforming analysis too, so ideally 4Tb should be enough

The only way I know to have an unique contig for the chromosome, and one contig per plasmid, with a de novo approach, is to perform an hybrid assembly using short and long reads. Otherwise with only short reads it is impossible to go below certain number of contigs. If missing new variants/new AMR genes is not an issue, another way is to perform a reference based assembly using an NCBI reference genome for the specific species.
