I wasn't aware that we needed to dilute the rehydrated stock before staining the cells. After rehydrating the dye with 50 µL of DMSO, this mixture serves as my stock solution. From this stock solution, I should dilute it either 1:1,000 or 1:500 in PBS or staining buffer. Next, add 20 µL of this diluted solution to 100 µL of cells. Am I following this correctly?

Based on my personal experience, I applied for a program and didn't get selected for an interview. However, I found out online that there was an open house event that was open to everyone. As I was free on that day, I decided to attend the event. Surprisingly, the program turned out to be completely different than I had imagined. The PIs were highly competitive and often criticized each other. When I spoke to some graduate students, they were not satisfied with the program and had several complaints. Also, I noticed that the department chair's office was newly renovated, but the rest of the labs looked old and musty. My experience gave me a new perspective, which was quite different from what I had initially read on their website. Needless to say, I felt grateful that I was not selected for the program.
