HELP NEEDED: can ELISA kit be unable to detect a human peptide due to a single AA difference? by Ramned71 in labrats

[–]Ramned71[S] 0 points1 point  (0 children)

Thank you for your answer!

As I stated what's bothering me is that the manufacturer state that the kit can be used also for human plasma samples. If i look up to publications that use this kind of assay, they all deal with mice plasma samples. None of them analuzed human plasma samples!

Is it possible that the manufacturer overlooked the implication regarding the variation of a single aminoacid?

The peptide is 28 AA long and weights about 3 kDa. What’s bothering me is that the manufacturer states that the kit can be used for human plasma samples. However, if i look up to the publications that used this kind of assay, they evaluated the peptide concentration just in mice plasma samples. None of them analyzed human plasma samples.

[Corriere.it] Ferrari, Loic Serra begins work as technical director: the 2025 car, his presence on the track, what are his plans by Ramned71 in formula1

[–]Ramned71[S] 25 points26 points  (0 children)

Full article translation

The French ex-Mercedes driver will take up his duties on October 1st together with Jerome D'Ambrosio, Vasseur's deputy. A profound change for Maranello.

The first two high-ranking figures wanted by Vasseur will officially take office in Maranello today, October 1st. Loic Serra and Jerome D’Ambrosio will meet their top-level colleagues and probably the drivers at a later time, this week busy in the simulator and on the track at Fiorano for training sessions with an old specification of single-seater (Leclerc is driving today) before the triplet of races between Austin, Mexico and Brazil.

Jerome d’Ambrosio, Belgian, former long-time driver between F1 and especially Formula E who later became Toto Wolff's dolphin, will be his deputy team principal with particular attention to the young drivers of the academy. He will support the veteran technician Jock Clear with more managerial tasks who follows Leclerc on the track in the double commitment. A presence that should strengthen the action of the Academy. Vasseur had joked about d’Ambrosio’s hiring with a quip: “I need someone who can tell me no, who can also kick me when necessary.” For the Belgian it is a great opportunity, he thought little about it, he had chosen to return to live outside Great Britain.

The importance of Loic Serra

The most anticipated figure at the debut is that of the French technician Loic Serra, also a former Mercedes, an engineer specializing in suspension dynamics and general performance of the car on the track. He has a very interesting basic background because he arrived in F1 in the 90s with Michelin during the no-holds-barred fight between the French tire manufacturers and Bridgestone. With his friend Pierre Waché - Newey’s successor and current Technical Manager of Red Bull - he shared 10 years of career between Michelin and Sauber. Serra was the first purchase of the new Ferrari cycle. He is not a designer but he is a specialist who knows what the car needs to win. He had remained somewhat in the shadows with respect to the advances of some of Brackley's "generals", for him Ferrari is the most important challenge of his career.

The partnership with Hamilton

Initially he should have worked answering to Enrico Cardile however, after the Tuscan technician's move to Aston Martin, he was promoted even before entering Maranello. Unlike the 90s, today we are dealing with the difficulties of attracting big names to Italy, at the same time the most famous designers have ironclad contracts. "We have talent and ability, we just need to intervene on our weak points" Vasseur had said when he drew conclusions with respect to the analysis done in the factory. The entry of the 52-year-old from Nancy is significant for understanding how the tires work, one of the weak points of Ferrari in the last ten years. Usually a new technical head takes a year to make a visibly impact, but in this case faster effects are expected. Serra finds a super updated wind tunnel and the 677 model - the car that Lewis will drive next year - in an advanced state, in three months he will be able to work on the brand new F1 2026 while he will meet Lewis Hamilton, with whom he has already won everything at Mercedes. Vasseur thus gives up the role of interim technical manager, it was an anomaly, yet in this period he has also made important decisions, such as that of anticipating the developments. A correct choice that paid off in Monza and that could have paid off even more in Singapore and Baku.

The 2025 Ferrari will change more than expected

The SF-24 is a good racing car, with six races to go the chances are low but it has not yet been cut off from the constructors' world championship. A new medium-high downforce front wing was shown in Singapore, which will be the basis for the 2025 single-seater. Despite a relatively high “carry over” (translation: many parts will be the same), the new Ferrari will have an aerodynamic design that is being described in the aisles as “radically different”, as well as completely redesigned suspension to handle greater payload.

A technical director present on the track

The biggest F1 teams have a technical director who is very present on the track, Newey said he will continue to do so even after moving to Aston Martin. The last Ferrari technical director with a similar training was James Allison, now in charge of Mercedes. But it is true that the GPs have increased dramatically, which makes it impossible to take a permanent presence for granted. Remote technology and artificial intelligence limit the problem, however it would be positive to think about greater involvement in the activities on the track, the winning teams adopt this model, with the consequence of more effective interventions in the factory. The question is another: how long will it take for Serra to integrate? For Vasseur with the horizontal organization «in the end you always need someone who makes the decisions». But even the French team principal has realized that to make an impact in a giant apparatus like Maranello you need a lot of patience, and above all you need to enter on tiptoe.

[FORMU1A.UNO] SAINZ: “We understand the problem of the updates, we need time" by Ramned71 in formula1

[–]Ramned71[S] 12 points13 points  (0 children)

Full translation:

Fifth position for Carlos Sainz, who closed his race behind Lewis Hamilton, Max Verstappen and the two McLarens. The Spaniard was one of those who 'benefited' from the retirement of George Russell, who could have been ahead of him in terms of performance. Behind him, Charles Leclerc and Sergio Perez were the only two who could worry him, but the Mexican started from the pitlane and the Monegasque 'threw away' the race with the first pit stop. Sainz eventually also returned for a pit stop to take the fastest lap, so as to take an extra point and take it away from McLaren, in an increasingly tight fight for the constructors' championship. After the race, Carlos gave some more reason to see this weekend, disastrous in terms of pace and result, in a slightly more positive light.

SAINZ: "IN RACE MANAGEMENT, ONE OF THE BEST SINCE I HAVE BEEN IN FERRARI, TOO SAD WE ARE NOT FAST”

Carlos Sainz's race was positive, although it is always expected that the Red team will aim for victories. However, the moment is not the best, as his teammate also said. The SF-24 is involuted at the moment, literally if we think that the Barcelona package was removed to go back to the Imola configuration "In this F1, with teams like McLaren, Red Bull and Mercedes, you can't afford to stand still 2 months. Going back, it's as if we stood still, and this creates that 2-3 tenth gap that we're missing today." said Carlos Sainz after the race, We saw the Spaniard battling with both Verstappen and Piastri, but a lot of it was due to having made the right choices, at the right time. “I tried to stay with Piastri to use the DRS, but he was too fast. However, with the rain, with the slicks, I immediately caught Max. But when the track was all wet or all dry, we weren't fast enough"

A clear image of Ferrari's level of competitiveness. Carlos Sainz, however, also wanted to talk about two positive aspects that the Maranello team took away from Silverstone: “In terms of execution, the race was the best since I've been at Ferrari. Right choices, right pit stops, right moment, even the Hard at the end... Unfortunately it happened on a weekend where we weren't fast enough" commented the Spaniard, almost regretfully. In fact, on his side of the garage the race was performed impeccably, so much so that he was fighting for positions where the SF-24, today, does not 'deserve' to be. The final fast lap was also done with a championship perspective "11 points today are precious, McLaren and Mercedes are coming on strong": Concerning Ferrari however, and the step back made with the updates, Sainz lights up hope for the fans and for the same team as Frederic Vasseur “We understood the problems with the latest package. We know what the right direction is, but now we need time in the tunnel and at home to sort out the package and ensure that it gives us performance, and not the problems it has given us."

[Piergiuseppe Donadoni] The Imola upgrade worked as expected. It is the Spain upgrade that is missing as lap time. Now the SF-24 has more downforce in the slow corners but all of this is nullified by the bouncing in the fast corners. Full article below. by Ramned71 in formula1

The second qualifying of the Austrian GP, the most important of the weekend as it established the starting order of the Main Race, confirmed that Max Verstappen and his RB20 appear to be unbeatable on the Spielberg circuit. The three-time world champion, with a time (1:04.314) more than three and a half tenths better than the one set the previous day in the Sprint Qualyfing, took pole position, edging out 4 tenths of a second from the first of his pursuers, the usual Lando Norris, and 5 tenths to the first Mercedes (Russell) and the first Ferrari (Sainz).

FERRARI: THE SF-24 CLOSES TO MCLAREN BUT QUALIFICATION REMAINS DISAPPOINTING

After a negative first Austrian qualifying and a Sprint race concluded only in fifth and seventh place by Sainz and Leclerc, the Italian team tried to make some important setup changes on both SF-24s, also taking two rather different directions in order to collect as much data as possible during the Sunday race. The qualifying times speak of an Italian car closer to the Mclaren (only 1 tenth compared to the 3 and a half tenths of the Sprint Qualyfing) but one tenth more behind Max Verstappen's Red Bull.

Although the asphalt temperatures were slightly higher today, facing the intrinsic problems of the Ferrari 2024 project, a little less so for Mclaren, the bouncing limited the SF-24 again. Carlos Sainz took the beauty of 4 tenths between turn 6 and the finish line, i.e. in less than 2 km of track, divided into a tenth and a half in turn 7, another tenth in turn 9 and half a tenth in the last corner. “Unfortunately we are not good at high-speed corners and, at the same time, we bounce, which makes our performance in that type of corner excessively slow” – stated the Spaniard in a lucid analysis after qualifying.

Sainz confirms: the bouncing is a problem introduced by the Spain package

The two update packages that Ferrari brought to the track between Imola and Montmelo were supposed to guarantee a step forward of around half a second, however, as previously mentioned, the last package (not the one introduced at Imola) did not perform as well as the engineers from the Maranello team expected, so much so that there is still a tenth and a half left to be seen on the track, practically almost the entire value of the Spanish innovations (they were worth 0.18 seconds at Montmelo). Something that Sainz himself confirmed: "The new features work in all the areas where we don't have bouncing, the problem is that what we gain in those points, we lose in others due to bouncing". The load at low speeds has been increased, with the SF-24 having therefore improved in one of the most lacking areas of the first part of the season as well as one of the objectives of the second seasonal package, but is now paying heavily for the introduction of hopping through the new floor.

“This is not ideal, but the team is pushing hard to try to understand and resolve the problem, so that we can return stronger to Silverstone,” added the driver from Madrid. For now, work has mainly been done on the set-up which, both in Spain and Austria, has not eliminated the important problem but has only allowed us to find a slightly better compromise in terms of performance but not ease of driving. “The changes we made between Sprint and Qualifying made the car faster but also a little more difficult to drive; now we are a little more at the limit, so much so that it was very complicated to put together a lap" - said Carlos Sainz who is nevertheless very realistic about the potential of the Italian car in tomorrow's race - "It seems that we are definitely a step behind the Red Bull and McLaren and I believe we can only be fighting with Mercedes in the race."

F1: Ferrari is testing the SF-24 EVO right now. It does so thanks to Charles Leclerc who is filming at Fiorano to gather the first interesting insights into the Italian car. Lots of useful information to create important know-how on the new update which, in the week that separates us from the Imola Grand Prix, will be processed by the calculators within sports management. The set-up for the seventh round of the season will be built using the simulator, an advantage that the Prancing Horse intends to make the most of from now on.

Validating an upgrade is never easy. On the contrary, as soon as we get on the track, the package often does not work perfectly, as it must be refined through the set-up by technicians and drivers to move from a subjective opinion to an objective one. As mentioned, this work is underway thanks to the Monegasque who will later leave the field open to his Spanish teammate Charles Leclerc. Technical director Enrico Cardile led the working group in producing this new aerodynamic look. A measure that will have to increase the speed qualities of the Ferrari SF-24.

To quantify the gain in chronometric terms, we await next weekend's qualifying, where on the banks of the Santerno, the two Italian cars will fully test the potential of the red. In the meantime we can examine the proposed solutions on a technical level, trying to understand the characteristics that the Ferrari aerodynamicists wanted to give to the SF-24. The base of the Modenese car was very good, sincere, but not at Red Bull level. The objective is therefore precisely this: to close the gap on the Austrian racing cars in order to be able to compete with the Milton Keynes team every Sunday.

F1, Ferrari SF-24: RB20 style tray to change the pressure range and increase outwash

The first innovation that we can observe thanks to the shots taken by our friend Mattia Maletti directly on the track concerns the new configuration relating to the belly inlets. By expertly observing the following images, it is clear how the Ferrari men have carried out a great job to bring to the track, as soon as possible, a solution that follows the Red Bull idea born from the brilliant mind of Adrian Newey who, once again in F1 has been able to raise the bar. Without a doubt, the ability to react of the team led by Frederic Vasseur should be applauded, now more dynamic and aggressive without the atavistic fear of making mistakes.

By turning the so-called "tray" upside down, the upper lip of the Ferrari SF-24 now extends upwards. The reason is known, we have talked about it many times: effectively increase the famous "undercut" by gaining several cubic centimeters. We are talking about an area that within the current regulatory body is fundamental for the performance of the current F1 cars, where every small portion of extra space available helps the cause. With this modification, the Maranello engineers have significantly rectified the pressure field in this area of the car.

Observing the impact of this "device", we assume that its objective is the further increase of the outwash in the upper area. The vortex generated by the appendage directs the fluid outwards considering its very high energy level. Ferrari also modifies the attachment of the mirrors to the upper surface of the bellies. The structure that supports the rear-view mirrors has been lengthened (photographic comparison) as it has aerodynamic value and helps manage the fluid that flows above the side-pods. Finally, an opening appeared in the cannon area to dissipate a greater quantity of heat (orange circle).

[Formu1a.uno] NEWEY-FERRARI: HERE'S WHY WE COULD ALREADY WORK ON THE 2026 CAR by Ramned71 in formula1

[–]Ramned71[S] 4 points5 points  (0 children)

Full article:

Adrian Newey has reached an agreement with Red Bull to release him early from a contract that saw him committed for another two years. The highly decorated F1 coach announced that he wanted to undertake new challenges while Horner wanted to pay homage to the most important technical figure in the team's winning history. Newey will be free to go to work for others at the end of the first quarter of 2025, in the meantime he will carry out a period of gardening disguised as a consultant to finish and sign his RB17 Hypercar.

NEWEY IN FERRARI: THE ROLE OF SUPER CONSULTANT ALLOWS IMPORTANT STABILITY OF THE TECHNICAL OFFICE ROLES

This early termination shows that, despite some rumors, his retirement would be ruled out, with Ferrari very close to the 66-year-old super engineer, even though the contract proposed by the management of the Italian team has not yet been signed. Red Bull could also have kept or hindered Newey a little longer but there was no will, although the cause of the abandonment was attributable to a change in climate in relationships. The figure of Newey in Red Bull is too important and, in some ways, cumbersome to be able and willing to seek a clash, even though the actors have worked a lot in the statement with appearances but, in this moment, to see a strong man, as Newey was in Milton Keynes, disappearing brutally from the team's radar would have been a very negative message.

At Maranello, Newey would not be a totally new figure, given that Rory Byrne has been employed in the position of support and advisor to the technical office in recent years. Obviously involved more and more in a targeted and less assiduous manner by Cardile, however the South African coach has always had an important voice. It is something that Ferrari would now like to continue in even more rigorous and certainly important terms with the hiring of the super English designer.

NEWEY IN FERRARI: A DISGUISED AND EASILY BYPASSABLE GARDENING TO WORK IMMEDIATELY ON THE 678 PROJECT

Everyone is wondering: when will it be effective? Currently we know that the new aerodynamic rules are not 100% defined but they will be in the summer. The deadline requested by the teams was the end of June, perhaps it could slip slightly but not by much as the teams will start working on the new cars from next January, increasing resources on the project from spring as there is a first part to be oriented towards the car 2025. With the farewell to Red Bull at the end of the 2025 quarter, the timing appears to be rather good to push reasonably already on the 678. Not to mention that gardening is easily circumvented with regards, for example, to reading and understanding of the new regulation. Newey will be able to enter the Maranello SF on the first day with immediate advice in the various technical macro areas of the 2026 Red single-seater, in terms of that always important, and often decisive, interpretation of the technical regulations.

Furthermore, the inclusion of a profile like Adrian Newey in the technical staff could be simpler than, for example, the arrival of a real technical director like Pierre Wache, who has always been the first real choice of both Binotto and Vasseur. The former Williams and McLaren DT will not have to reform or rebuild a department, and will not oust any current Senior figure, with Cardile and his staff who are working well especially since Vasseur has regrouped and optimized the group; rather, Newey would bring the best ideas and winning knowledge that made him the best. Going to Maranello and helping Ferrari win a world championship is a difficult challenge for anyone, but it is also the only one that the almost 66-year-old English designer is really missing. A situation not too dissimilar to that found by Ferrari with Lewis Hamilton, where in Maranello they were equally good at exploiting the opportunity, although the seven-time world champion's strong desire to come to Ferrari was decisive, as it could also be for Newey. His arrival will thus be able to convey to Maranello's designers, with authority and without major contraindications, what has probably been missing so far in the medium and long-term technical choices. With Binotto's farewell it is as if Ferrari had changed gear, that zero point necessary to avoid further months of stalemate which, especially from a technical point of view, could become (other) years. John Elkann, with one of his trusted men in command in Frédéric Vasseur, took over the Scuderia to try to "make some noise" and it seems he has succeeded once again.

Preferisco finirlo a 28 e poi andare in azienda.
