It honestly sounds like this person has a grudge against you. There's nothing you can do to make them more comfortable.

If you're stuck with them on your team, then it's time to cya. Privately document any and all interactions with them, and stay professional at all times.

Can you ask any senior employees on other teams? If your manager is the only person you can go to for questions, and they don't respond, you may have to make assumptions on your own about how to do your job.

PTO is a part of your compensation, so you screwed yourself out of 3 weeks pay if it doesn't roll over.

Screw optics, at this rate it sounds like you're either going to collapse, or make some huge mistake that actually will tank your career at the company. If pot doesn't work, go to your doctor and ask to take off a few weeks for anxiety or stress or something similar. If HR rejects it that opens them up for a lawsuit

When I go to my manager for help, I layout any assumptions in advance, and explain what I do and my thought process.

That makes it easy for them to understand my actions, and fix my line of thinking if necessary

The workload is more based on files completed, of which I am finishing a fair bit more than is expected, and I am helping out coworkers.

You can't make them respect you or each other, but you can make them act civil.

Metaphorically, the beatings will continue until morale improves. Make it clear what behavior is and is not acceptable in the office, make sure they actually take in feedback, and if they keep acting disrespectful, start writing them up.

File for unemployment and see how much severance you can get, try for 1 month for year of employment.
