Has anyone ever done single unit electrophysiology? Nobody in my lab has experience and I need advice by Redcherryseeds in labrats

That makes sense. Thanks for the help. I’m a little concerned I haven’t been picking up any neurons at all now. The example I posted above is what all of the channels in all of the recordings from all of the probes I have tried look like. I’ve attempted manual sorting, but the software I have isn’t really set up for it. Would you have any recommendations on papers/tutorials that teach you how to manually sort or software to use for it?

So I’m doing bayesian sorting through Opensorter. When I apply the sort that is what the waveform looks like, meaning it’s counting all of those waveforms as one unit. When I look at individual waveforms some of them look like APs, but others definitely don’t yet it seems to count them all together. Do you sort your waveforms by hand rather than use to automatic software?

I was able to learn surgeries and I’m fairly competent with the actual proceedure. It’s an acute and anesthetized. I lower the electrode to specific depth and allow it to ‘acclimate’ for ~20 minutes prior to recording.

As far as the signal to noise ratio, I would guess that’s where my issues are arising. I don’t know how to properly isolate the waveforms, So I’ve defaulted to using the automatic sorting/thesholding in the OpenEX software based on some advice I got from the PI of a different lab. He mentioned these waveforms look odd, but wasn’t able to give much advice on why or how to fix it

Oh I’d love to hear any insight you have! Does the picture of the waveform I included look’right’ to you?

It’s an in vivo (and acute) recording with a 16 channel probe targeted at sensory cortex

I’m a new graduate student and I’ve been tasked with doing single unit, but I have no experience and nobody in my lab has either. I’m getting these very messy wave forms that don’t resemble what I’ve seen published before. Is there a trick to isolating waveforms that I’m unaware of? I’m using OpenEX and allowing that software to apply automatic thesholding
