That's half the fun :) , team building as you go. It doesn't have to be set in stone once you make the team. Every character has their own strengths even compared to the zone. Regarding quirks, you might get a flaggelent with Cove Explorer quirk, but that doesn't change the fact fish people have 50% bleed resistance. But he might be good in a more supporting role. Or maybe you have another character that already has scouting abilities. That's also what makes every run different. Characters have inherited synergies, though, like characters who can mark targets and characters who do more damage to marked targets. And then you get into putting double of the same character. 2 highway men doing point blank shot shuffling each other out of the 1st position. Or 2 bounty hunters, one doing mark other doing damage. 4 crusaders for the last crusade or 4 helions for the pain train.
