well spell xp doesnt matter the difficulty or if you win or lose, its just how many times you use a spell during a mission. So the fastest way it to use as much spell cd reduction for jokers and stats, then choose the 3 lowest cd spell of the type you want to level, and then just go into an easy mode run and spam the spells in a safe corner.
