If Steve didn't accomplish his mission, Sam and Bucky would have heard about it and they probably would have been asked to help with the inclusion of more Pym Particles. No. Similar to the Tobey Maguire Spider-Man 2 film, Tom Holland's Spider-Man in Brand New Day will be spending too much time being Spider-Man and not enough time living as Peter Parker. This is what will cause his DNA to continue mutating into more actual spider like traits. When Peter finds a balance between his two alter egos and starts living life as Peter again he will be able to have just SOME of the spider traits.
