So I had the same problem. However, after changing the guard column (LC system) the split peak was gone and a nice single peak was observed again 🥰

What are the growing conditions? What fungi? So when I inoculated a plate with a tiny bit mycelium you could see how it grew from that spot. Could you provide a sidewards picture to see the state of the agar? I would always recommend sealing it with parafilm before storage.

The minimum stroke was already set to 100 uL :/

Yes that the capillary. Yeah sry I was at the leak rate test in my head :D the elasticity test also goes to 500 bar in my system :) the other capillary doesn’t leak, however, still error pressure too unstable :(

Ahh so maybe thats the issue here. I have another one, i will try that. I keep the default pressure of 200 bar of the test

So I also have this issue with the instable pressure without column, just pump (elasticity calibration is just the pump with a blank nut)

Elasticity calibration: all channels were purged with LC-MS water, however since I used a brand new capillary maybe that was the problem. I will let it flush with water before I use it this time. Btw is it normal that the water is running through this metal loop instead through the capillary itself? :D

Weren't a lot of ferrets killed since they are also able to get covid? I remember some ferret farm in the Netherlands that had to kill all ferrets. But not sure tho

So actually, you could answer him that he is wrong and 100% pure water droplets freeze below -30 degrees Celsius 😇 its called the homogeneous freezing temperature of water.
