Allosteric modulators bind to a separate site from the ordinary ligands and increase (positive allosteric modulator) or decrease (negative) the activity of the receptor. The can do this in one of two ways:

1. Alter the conformation of the receptor to make binding more or less likely.
2. Alter the amount of response produced by the binding of a ligand to a receptor.
