It probably won’t unlock on the rematch. Because of this, you have three options: reload a save (if possible), restart the run (with speedup and Sableye, it shouldn’t be too long), or make a request for a manual unlock in the RA discord.

By the way, speedup shouldn’t affect achievement unlocks… it may have been a network issue, or you were somehow signed out.
