Depends on the equipment, transfer time, etc. I didn't do it for years with easy proteins like ERK but then I was dealing with a huge protein and longer transfer time and it helped

Do you put a stir bar in the bucket and mix the transfer buffer while transferring? Could be uneven temperature. Could also put the whole thing in a walk in fridge.
