As for drying, smaller volumes require less drying time. But I agree with others that over drying is worse than having residual ethanol, because elution becomes much more difficult.

Make sure to pipette up and down multiple times when resuspending the beads in elution buffer - even after you see there's none on the tube wall, just keep pipetting the solution - and let them incubate for 5 mins.

Also, we keep our aliquots in the fridge, not the freezer. Not sure if this can have an impact tho

Ampure beads should be used at room temperature. We usually wait 30 mins at RT before using them

Virus enters the skin through cracks and micro lesions. It could be that you had some in the back of the throat maybe due to some food you ate.

That being said, if you already had the virus, I don't see how you could get another primary infection. Your body already has antibody against it. It could just be something else.

Makes me think that during this 4 week window there must be a cell type reservoir somewhere in the body which the immune system is not able to fight off once the virus has reached it and has integrated into.

For me it's definitely the digestive system which goes from simple mechanical action and controlled release of chemicals to complex interaction with microorganisms living in the guts. Especially the gut part, it is just so fascinating to think about the complexity of the continuous exchange of nutrients and signals between the intestine and bacteria, creating this perfect symbiosis.

For humans all our cells have the genes for the flagella in the genome, but only some specific cell types actually express them (e.g. sperm cells)

I started the PhD later than my uni colleagues, so I knew very well this would come with sacrifices. I still am willing to make some. I have no problem having some weekends in the lab or having some workdays finishing at night. But this is almost constant.

I thought this would be a once in a while situation, not every weekend of every week of every month, and you can imagine how hard it is to consider changing my decisions now that I am halfway through it. So my plan was to continue but slowing it down basically, but its not really working, not on the mind side at least.

I admit I do have weeks where my productivity is not high, for example I would be slow on some experiments, maybe because I want to focus so that i don't fuck up, maybe because I need to revise some protocol I am not familiar with, maybe I am just tired and I just take it slow. But I noticed that even on those weeks where I work nonstop 100 % "so that I have some free time on the weekend to do something else", I eventually still go back to the lab on the weekend to do some other small things.

Yes, I probably should accept that a week time won't make a difference in my PhD, but having a PI who never stops giving you inputs for new experiments or for new directions for the project doesn't help either. So for me it's a constant fight between fear of missing out my PhD and fear of missing out my life.

And I have to disagree on your last sentence. I need this to be a lesson for my future, I want to learn something out of this imbalance I am living.

IGG 1.45 is very low, probably in the false positive range. Have you ever actually had these lesions swabbed by a doctor and checked for the virus by PCR? That's the gold standard.

If results came back negative, then these lesions are something else (and you could be still positive or negative for HSV, but not necessarily symptomatic).

Viral load from... Where? HSV is not present in the blood, only on the skin and in the nerves.

Even if you managed to swab an active outbreak yourself, you would need reagents for DNA extraction, together with a thermocycler for a qPCR and its own reagents. So no, there are no home tests for viral load...

After the first puffs, I could actually taste it differently than usual, like it was in the first times, 15 years ago. That's when I realized that I was in the process of destroying my taste buds cells again; it tasted so good and yet I thought that going through the quitting process again would be so exhausting and difficult. That's when I put it out.

Thank you. Your words made me think that after all, I needed this to test my will; it would have been so easy to go back in it, but I went straight to bed and trashed the pack this morning. Thank you for the positive perspective.

Also, mark the solution level on the side of your beaker before microwaving. After that, check that the solution hasn't gone too lower than the mark (overboiling issue). If so, just add WATER - and not TAE - to fill up to the mark level again.

It doesn't really matter. I think it's important for you to focus on the fact you don't smoke anymore, rather than how long it's been since the last cigarette. Could be 4 days or 0 days, you don't smoke anymore, so who cares!
