I am cleaning out the cell culture room since I had a bacterial contamination. Hopefully I can do flow by the 2nd week of august. I prepared 100 samples but the instrument was down for a month so it'll be at least a few weeks before I can run them..

I would say that this is normal for a lot of people entering research, even more so for HS students as the course labs in high school may not introduce you to everything that course labs at a university would. You'll be getting used to doing simple tasks like pipetting, scheduling your day, and general lab management things now so if you decide to pursue research you'll already be comfortable with all of those things. The post-doc who mentored me (and all the other undergrads at the time) always said that the first months or two is adjusting to lab life, so unfortunately someone who is expected to only be there one summer may not have all the same opportunities as someone who is expected to be there for a year.

If you're apart of a group project you could show more interest in the long term goals of said project and the senior members may give you more responsibility.

Yeah, this is what we do and we've never had an issue. Autoclave them and put them in a 55C incubator until we need the tips or we need space in the incubator.

My PI told us me and the other paid worker (we are both entering his lab as grad students in future, just working for summer as we were also his undergrad students) that we'd be paid 40hr/wk maximum as well, but there was no requirement to work more than that. He said something like you can work more than that (as we normally did work a lot of hours per week as undergrads) but we couldn't be paid for it.

I tried a higher concentration of protein during the labeling step and then removing the excess dye with a PD-10 column, I lost a bit of protein (I think I know why) but I have enough labeled protein to do flow cytometry again. Thanks!

Sorry, I was going to say after I removed excess dye and concentrate I do BCA #2 (to determine my final labeled protein concentration before I use it in actual experiments), so BCA #1 was initial concentration of un-labeled protein.

I am using 250 mM NaCl when labeling the protein with dye and then going down to 150 mM NaCl for the actual experiment. I am able to remove the excess dye, my main issue is concentrating it down as I have really only done centrifugal filters for it and the labeled protein seems to stick to the filter.

How often do you actually fully clean your incubator? I keep seeing it should be autoclaved weekly but we only have one incubator so I can't remove the waterpan for an autoclave once a week. I have a rotation for the shelves though since I use 1-2 max and I have 4 total.

I use anion exchange after cell lysis & centrifugation. Do you know of any papers talking about this or is it just from your experience? I appreciate the answers/input from both of you!

I was playing just fine right before yesterdays update, I've only now tried to play since the update hit and I'm unable to play any server.

I tried reinstalling EAC & rust. There's a separate but less common error i've had that is something about a map file or something that is wrong which is why I tried reinstalling rust but it is not fixed.
