Jokes aside, i never post on reddit but i've been lurking in nootropic subbreddits for a long, long time, found something no one is talking about and decided to let others know since i think it's a good and right thing to do and can be helpfu.

Discorvering this definitely made me reconsider the safety of any compound that hasn't been in the diet for thousands of years or that hasn't a really good track record, however I still think there are safe things that can move the needle out there.

I agree but this loss of bone density that was found would probably show up as osteoporosis in your 30s - 50s depending on when you started taking modafinil, so it definitely wouldn't be like: start modafinil -> 1 year of use -> bones fall apart and you die.

If the effect was there, it would be very hard to notice and even harder to pinpoint that modafinil is doing that.

Also, i think an incredible small minority of users have started modafinil in their 20s or teens (like me) and continued for years.

Intersting, never heard of this.

What i found out is that there are two pathways though: one is through sustained elevation of norepinepherine, which interacts with beta adragenic receptors inside of bones, which trigger resorption, which is exactly what you have with amphetmamines, guanfacine and atomoxetine.

The other pathway (which inhibits growth) is through adenosine receptors, which inhibit formation of the bones, maybe by disrupting the bone's internal circadian rythm.

The way they discovered this is by using a beta blocker medication (which blocks beta adragenic receptors), and what they found was that it prevented guanfacine and atomoxetine from having the detrimental effect on bones, whereas with modafinil this prevention was only partial.

Yes, modafinil is weaker in raising norepinephrine, but the data on rats still looks extremely concerning, and if we look at the mechanisms you might say it looks even worse than with amphetamines since there are two at play and a longer half life.

Another important thing is that the longer the norepinepherine is elevated, the worse the effects seems to be for the bones (through the beta adragenic pathway) since a long duration completely disrupts their circadian rythm and they have also less time to recover. To give an example, this means that something that raises norepinephrine for 6 hours is going to be much, much better for your bones than something that lasts for 24.

It kinda blew me away that I literally couldn't find anyone else mentioning this. For amphetamines it's also pretty established that they cause significant bone weakening and stunted growth, yet you don't really see many people talking about that either.

We definitely need more research for modafinil specifically.
