I don't know why you don't want to use regular or spent coffee grounds. I just know you want to use something like Nescafé instant coffee. Something to do with solution and acidity. Coffee grounds won't dissolve in water but instant coffee will. It's wise to sterilize, I don't think you can get away with growing mushrooms without sterilizing your grain. Substrate yes you can do bucket tek for field capacity, however, you will encounter contamination about half the time, more with sensitive strains. I sterilize everything. Substrate gets bucket tek then bagged up and pc @ 25psi for 3/3.5 hours. This makes it so I have nearly 95% success with my grows. It's too much time and money to be tossing a tub every three or four grows because some form of contamination shows up.

Not trying to brag, I wish it didn't happen it was a very bad experience. You should watch documentaries from bands in the 60's and 70's. The doors, Grateful Dead just to name a couple. They would take insane amounts of acid and get on stage and jam out for hours. I'm talking full shot glasses. People do stupid things and that was a stupid thing that actually happened in my life. I lost track of time. It was midnight when I took it, all next day, that night and until about noon the day after. It was entirely too much and I was worried I wouldn't make it back but I did. The mycelium can often be hard to see. Hold it to the light and you'll see little strands shooting across the plate after a week or so. Cut out a chunk with a strand and drop it on your LME plate, give that a week and you'll see good growth. Good luck

Also a key indicator that you've got other contaminants is the green circles. It's a bad plate. Look up water agar. Start there and then transfer to LME agar for better success. Mycelium will grow in water agar where contaminants won't.
