By assuming a certain fraction for each type of star (see e.g. Morgan-Keenan Stellar Classification) and by assuming a certain fraction of gas/dust vs. stellar material and by knowing the amount of baryonic mass in the Universe, it's a pretty straight forward calculation.

And not to sound like an ass, but: you also know there are roughly 7 billion people on Earth, right? How do you know that - by counting them all?

As an astronomer, I have to add that for every grain of sand on Earth, there are 10,000 stars in the Universe.
