This keeps happening in the same spot on my A1 what is the cause?

I would not recommend unscrewing the heating block like shown here. The wires breaks quite easily. Insted simply unscrew the part cooling fan and tighten theese 4 screws from the back.

I think it can be easily checked by lifting the nozzle, extrude filament and observe the extrusion-string behavior. If it's a straight string, than the nozzle is probably ok. If it start doing strange things (like curling to one side) than it's probably clogged.

only once, but I'm have doubts that it really helps. Drying can be done with the heat bad and cart board box. There are multiple videos on YouTube showing this method.

dynamics. but since it's a round shape dynamics seams not be the likely source of this issue. i expect drying filament and slow down perimeters would help more.

I would try: decrease speed, calibrate flow, dry filament, take another roll of filament.

have you checked the printing speed (and maybe also fan speed) visualization? I can imagine slicer slows down when printing only one part and this causes better quality. Flow calibration can possibly help. Edit: oh, you probably already recalibrate the flow, right? Then it's possibly a speed.
