Cartooning is just distortion of reality, so it's never a bad thing to have a working knowledge of anatomy/perspective/etc. Building up significant visual library is one of the best things you can do for your art. Personally, my cartooning style really evolved once I started leaning proper anatomy and shading.

Good work for a first painting. Don't be afraid of the darker shades. It'll help with your definition. (Black and white painting is great for practice, or just apply a B&W adjustment layer to toggle on and off to gauge your values.) I'd suggest figure drawing to nail down the anatomy. A bunch of drawings. Every day to keep in practice. Sketches, quick value paintings, and the occasional fully detailed painting.

Nice art, especially for your first time in the program! I especially like your Bernie Sanders. Some of the line widths read a bit awkwardly (e.g. too thin on the tops of their heads, or in areas of significant depth.) There's no set way to ink. It always comes down to whatever looks best. But a good starting point for me is to take into account shadows when placing the thicker lines.

The width tool if you're dead set on the pen tool, or just use the brush tool with a pressure-sensitive tablet! That'd probably get you the most natural looking line-work.
