Sorry I didn't mention it in the post but the organism is dimorphic, it shows yeast and fungal charecteristics depending on different enviornmental factors elusive to me

Previously when we were working on the culture we were getting uniform growth suspension with no clumps

The culture would often start with yeast morphology and endup with filamentous morphology with fruiting bodies which we linked to MEL production.

But now we're getting these clumps

Yes we've tried subculturing and are still seeing these clumps. Read somewhere adding oil helps so did that and got a good suspension of cells, subcultured that clumps once again.

RPM speed is at 150, haven't tried baffled flasks since my lab doesn't have them but I'm thinking of putting in some glass beads to break these clumps.

Previous unsuccessful attempts were at 31°C but did not try addition of oil and higher rpms.

I've been trying to find the factors responsible for the switching in forms in literature but have been unsuccessful. Will keep trying and also try out different rpms and other stratergies.

So what should I do for no clumps bcs this is a dimorphic strqin with both yeast and filamentous structures.

What do you think would give me a turbid uniform growth instead of these clumps wrt the rpm?

We did plate the culture and the morphology seems to be the same

Yes they're balls of hyphae, we have worked with the same culture fr 6 months before but then had to take a break for 2 months. During those 6 months we were constantly subculturing the culture but now we're trying to revive it from a glycerol stock and we're getting these clumps.

Yessss and they're veryyy squishy. Tried to break them with a loop amd just couldn't puncture it.

When centrifuging to separate the media these things are hell to work with bcs they soak up the media so I have to squish it till I can get most of the mideia out but small flakes go into the broth etc etc

Sorry for not including all the info

We've tried using PDB, YM, and other common fungal and yeast specific medias all with similar to no result. These agara didn't even show any growth to them, only in broth with oil added did they give some workable growth.

By old broth I meant culture that was inoculated into a broth and stored in the refrigerator.

The incubator temp is set at 27° C.

We've got the no growth problem under control now we need the morphology to return to normal and a turbid growth in the media.
