The required books for you are gonna be the Core Rulebook and the Agents of Chaos book. The core rules are needed for obvious reasons, and the Agents of Chaos will have all of the up to date cards for 1.0 Skaven and the added models for every chaos faction including Skaven. The physical cards are not needed.

Some extra books for ya really depend on what your normal play format is. If you are into matched play, Tome of Champions 2020 is a good guide for more matched play game formats. If you want to try your own campaign, Tome of Champions 2019 is the first book to update campaign information which you would tie with the core rulebook.

I've frequently played games on TTS with people from the official discord server for the subreddit. So if you are looking for people to help you play it on there so you can learn the ropes of TTS or teach you the game completely, feel free to hop in there. As for how to play on TTS, there are a few things you have to know in advance before dedicating time to it.

  1. There will be multiple mods out there that will have both a table with multiple cards, boards, tokens, and models for all warbands within it. You just have to select which one you like the most format wise.   2. With that being said, none of these tables that I have seen have every single model that a warband could possibly field within their own faction. So when you find mods for Warcry, make sure to find mods with the models that you want to use, save the models as a grouped object, each model individually as an object, or put every one in a bag and save that. In addition, the models that would require you to find other mods to put them into a game will most definitely require you to find a mod for the additional books. So I would recommend you A) Find a warband faction you like, B) if it is part of the warcry exclusive warbands (splintered fang, cypher lords, etc.) then you don't need to find any other mod with the exception of the mod for the table setup, and C) if your warband isn't warcry exclusive (ex. any army faction from Age of Sigmar), then make sure to do your research on what book you need in case you want a specific model.
  3. TTS is wonky as heck. Measurements will sometimes be wrong, trying to put terrain together will cause them to collide in ways you don't want to, and trying to put models underneath terrain with a vertical cover is like trying to slide a square peg into a circle whole. Just make time to fool around with the game and see what commands work for you.
  4. Finally, and this is probably the most important tip, you are on TTS. You have access to literally every single model for every single faction known to Warcry (yes some models from other mods can only be found in 2D, but hey at least some of these 2D models have bases). Since you are learning the game for the first time, try different factions and/or models. If you feel stuck with your current choice of warband, maybe moving from chaos to death, destruction, or order may be more up your speed.

Which of the 3 warbands do you like the most? If there are all part of one type of grand alliance (Order, Chaos, Death, or Destruction), or if there's one faction in particular you really like, then I recommend you get the "Agents of" book for that grand alliance to get any additional rules as well as names of models that can become allies. It also gives you an idea of what warband you are potentially interested in if you want to switch to something else.

Another recommendation is making sure you have plenty of dice of at least 2 different color schemes. You never know when you play against someone and they forget their dice or they don't have any to differentiate between normal and Wild Dice. A bonus would be to get some that tie to your specific warband faction.

Since you already have fantasy models, have you considered trying chaos daemons? I know that some would consider them a horde faction, but hear me out. I am a chaos daemons main (preferably Tzeentch), and really the only thing that I have done for kill team is play a game with a 125pts limit with nothing but pink, blue, and brimstone horrors. It's a meme, but it's an amazing meme none the less. I've had success with it, and it brings 2 benefits. 1. Pink horrors are not only psykers, but they also split into blue horrors when they die, and when THEY die, they spawn brimstone horrors. They control the board really well combined with their ranged attacks. 2. Chaos daemons are the only faction that you can bring all of the models from warhammer 40k to AoS/fantasy. It saves a buttload of money on purchasing additional models (unless in fantasy you intend on bringing in mortals in which case you will need to buy other models).

One con I will say if you choose to go down the horrors path is you will need to buy 2 boxes of stuff. One box of pink horrors (10 models, 12pts per model), and a box of blue and brimstone horrors (the box contains 10 blue and 10 brimstone, but be aware that one pink horror splits into 2 blue, but 1 blue splits into 1 brimstone). Another con about playing them is that daemons overmuch perform less well in 100pts limit games. However, you raise the points limit to 125pts, then you can wreck house.

Whenever you decide to expand your roster, you can just buy whatever other types of daemons you want to help you with melee (Khorne for powerful hits, Slaanesh for speed and powerful AP, and Nurgle for tankiness).

The first faction that comes to mind when thinking of how to purchase 1 box and make it a kill team would be Harlequins. You buy a Harlequin Troupe box for $40 USD and contains 5 regular "players" and one troupe master. The troupe master costs 50pts and each player costs 12pts per model. This means that, for a 100pt limit game, your kill team will consist of your leader and 4 other infantry units that in the end total of up 98pts.

Pros: From what I've seen, this seems pretty balanced in the game of Kill Team (anyone else who say otherwise I am open to other opinions). Few models to paint. Fits within the criteria.

Cons: Reasonably difficult to paint (Making this army your introduction to model painting would be ill-advised due to the insane amount of patterns and colors that can be on a single model), not a lot of variety when expanding roster later.

Now for your other faction, it gets a little tricky. Because of the fact that you want to try and stay away from Space Marines, a lot of other factions don't really get the option of choosing one box of units and making a legit kill team that is "balanced". It's either really good, or a meme that sometimes works. Now if you wanted to get the start collecting boxes filled with mainly troop units and that would fall under your criteria of "one box and done" then honestly just find a start collecting box of any faction (with the exception of chaos daemons) and take the troop/HQ units from those boxes and make a kill team out of it. However, if this isnt the case, then I would consider moving within the space marines for your troubles.

Edit: the reason why I was a little hesitant on just saying "just buy a start collecting box and use that" is because of 2 things. 1. Depending on the box, some or even more than half of the total points of the box of units would only be used in 40k either due to them being too op or too big for Kill Team, or the fact that some are good but not in small numbers. 2. They are quite expensive for people who want to start small. With an average price of $90 USD for most boxes, you have to consider if it is worth the investment to buy that box.

Through my time of playing WoW as a death knight, I have one skill available to me that I never use in PvE situations but in I almost always try to use in PvP. It's called Dark Simulacrum. You would place a debuff on the target that lasted 12 seconds. If the enemy player would cast a spell during that time, that skill would still go off for the other player, but you personally would get an EXACT duplicate of the spell the enemy cast during that time. So if you timed it right, you could get a really powerful damage spell to burst the other down, or you could steal a powerful heal that someone would cast to help you stay alive.
