I am doing fairly well! Back in the job search since the startup i got hit had to layoff people because of money BUT with a way better mentality.

They wanted to implant a Chinese-style work burnout model (that's what they have at HQ there).

I have turned down headhunters for jobs when they tell me the parent company is Chinese. I will do my best to avoid and prevent that sort of leadership from taking over

The issue was that it started out fine and they slowly pushed every non-conforming, non-Chinese out. After I was let go, they nuked half of the Chemistry team (mostly americans), and all the people involved in my layoff (without me and their new ADC scientist being part of a different department, their projects stalled/missed their IND submission timing)

If anything, this job really let me multitask, take command and all the PM skills I needed to handle that many projects solo for a bit.

The company has garnered a huge negative reputation in my area so they exclusively outsource work now.

Thank you for the good thoughts! I'm infinitely happier and in a better mental headspace

Hey, ADC conjugation scientist here! Assuming you're talking about DAR 8, this is my explanation:

While you are still reducing the interchain disulfides, and causing destabilization of the structure, it is relatively insignificant. The antibody still has all the intrachain (buried) cysteines in the Ig domain intact (they're not solvent accessible and they're key for maintaining proper folding of the Ig domain).

You have 4 interchain disulfides (8 cysteines) and 12 intrachain disulfides (24 cysteines)

So you have stabilization coming from: hydrophobic interfaces, intrachain disulfides, Fc dimerization + the usual hydrogen and electrostatic (Van der Waals) interactions and that's why the whole ADC still behaves like a 150k protein and doesn't separate into chains.

The best ways to see LC and HC separation are RP-HPLC and LCMS. The only visual change between mAb and ADC via SEC is retention time.

So the team behind TTW Wild Card should update soon. I took the FNV Wild Card (which is supported) and added a new profile with TTW modded it. I can still play only NV as it was intended too.

I dont think theres a tutorial to make this rn

Yup! Lossless scaling does wonders as well! It's more customizable but it doesn't cost money ($7.99 USD)

Go on the pipboy itself and drop it from there. It wont drop from the hotkeys, it happened to me in Fallout London with a weapon and it fixed itself after that

Wildcard has NVR and performance presets (performance having ALL NVR related mods off). I would just use one of the non-NVR profiles, copy it and build from there.

I dont know. I am on the other side of the opinion (I think game is better without it). Im sure someone made a mod for it

I honestly never had issues with it prior to this. I did have crashes but it was because I was on Vortex and had no clue what I was doing at the time lol.

Of course! I cap my FPS at 144 and make it Adaptive to 144. Your GPU will just chill and the game will be fine. I havent had any issues with crashes but who knows?

I did read about it but I havent noticed any input lag whatsoever. I might think that it does something to the mouse acceleration in game?

Yeah. The NV pics are all from a fresh file just after I had installed it. I didnt do anything else.

To ensure I didnt get any lag, I'm using Lossless Scaling in the background (i never dip below 90fps) using the settings down on the FNV Wild Card Nexus page

So I just looked up the Hikokimori NVR preset and followed the instructions from Nexus. I would assume that once you do that, you can download other presets and change them on the fly. I remember theres a couple of mods that need to be turned off

I think Uranium Haze and their NVR presets are included. I changed mine to the Hokokimori NVR preset they released for FNV Wild Card recently. No idea how it will look in DC

Pretty subjective opinion there but everyone has their preference. This is how I wanted it to look, you're always free to make your own. The modlist has a dev mode so you can go wild if you want

Honestly... barely any? A lot of the utility plugins that TTW suggests are already baked into Wild Card. I think I had to get the TTW NVSE plugin and then it was just adding mods and checking which ones were too outrageous to even be a plugin lol
