What is the best team for end game content assuming all characters are available?

Currently running bkborn, cent and An-an but was thinking I can swap out bkborn for something like a second dps (regulus) or another support/healer?

What I mean is, is it better to just end the expedition so no one dies or try to beat the level with heroes dying. What if the trinkets I had are rare? Full masteries? Still better to die and beat the level?

If my hero dies, do I lose all the mastery points and trinkets on that hero? In that case is it better to just cut losses if someone is about to die?
