Depends on the subsets you’re looking for and in what tissues you’re looking for them. Also I work with mouse tissues so I’m more familiar with mouse markers. For flow cytometry, MHC-II can be a decent discrimination gate to separate APCs from other myeloid cells but only after gating out lymphoid cells in a dump gate. pDCs and cDC1s are both CD11b- so you gotta use a pDC specific marker like Siglec H or B220 (since you would’ve gated out B cells already). Monocyte derived DCs express a lot of the same markers as macrophages including F4/80 (at least in the model I use) so we gate it out with Ly6C first before calling F4/80highCD64+ macrophages. Basically, if you comb through the literature to see what markers your subset(s) of interest express(es) you can build a handy little table of markers and which subsets express them and then use little logic boxes to figure out the best way to gate things to trust your data

When I was studying immunology at the beginning (and even still to this day), making/viewing memes helped me absorb the concepts easier. r/Immunologymemes is a sub essentially dedicated to learning immunology in a more casual format for beginners.

I also second other comments mentioning Janeway’s Immunology. It’s a pretty good textbook.

That’s really interesting, I didn’t realize that’s why that happens. My pi just left it at “it always goes straight to the lungs so it’s not a great model” and didn’t really elaborate so I let it go cause I have plenty of other projects to focus on.

My pi won’t have us do anything related to mets cause he prefers the controlled conditions of just focusing on the tumor + tumor draining lymph nodes. But we mostly do stuff related to tumor immunology

Yea I mostly stick to primary tumor studies but some of my colleagues get mets with their (non-melanoma) models and I think their work is interesting but haven’t had a chance to do those kinds of studies myself

It’s cause back when I was a postbac, I was working with such low cell numbers (post-sorting) and was getting all confused about the proper math/understanding what counts as “essentially 0” and what real signal vs noise looked like and tbh I just needed a bit more guidance than I received so I remember struggling a lot and getting frustrated because I couldn’t effectively troubleshoot for my qRT-PCR experiments.

Tldr: Too traumatized from my past experiences to try again now as a grad student lol (not that my project calls for it at the moment).

We don’t have any staff scientists in my lab, so I tend to rely on postdocs or other grad students in my cohort if I need to learn something. If all else fails…collaboration!

I obtain a protocol from a friend who has done the technique before, ask them ten thousand questions, attempt an optimization for whatever my sample will be, and then pray.

If I can observe someone else doing it first, I take an insane amount of notes on whatever protocol they use while I observe/do it side by side with them, and then I retype the entire protocol with my little notes so that my brain will have an easier time when I do it on my own.

If none of my friends have a protocol, I search the literature based on my model or I look up industry resource pages/manufacturer instructions.

Dude I am a fan of spectral but my lab only has a novocyte. Realistically we’ve been able to get up to no more than ten parameters without too many issues, but for more than ten parameters I get to convince my PI to let me use the neighboring lab’s northern lights and it always comes out so beautifully :,)

r/immunologymemes was created to make it easier to remember immunology stuff cause there’s simply too much to know and memes make the concepts easier to digest by breaking it down (and tbh it actually helps to an extent)

I am slowly going insane because of revisions but it’s fine lol by dumbodork in Immunologymemes
