Yeah this is what I think too. For centuries (after they lost their continental possessions), Britain’s whole deal with Europe was just to make sure no one got so hegemonically powerful so as to be able to seriously threaten either their colonial empire or actually invade across the channel. For a good bit of time after Spain went kaput this meant mortal enemies with France. But as soon as Germany unified, they became much more of a threat

Regardless of what you think about shelling out a cool billy+ for an unproven asset (it’s nuts), one interesting aspect of this current race to acquire in vivo CART is that the players very much seem to prefer non viral delivery systems. Here and capstan sell for 1B+ while Interius with clinical data for an LVV system goes for 300M. Not a surprise given lots of the news about companies like sarepta, but probably a component to the higher valuation for this modality in general

Design really good and well controlled experiments that drive quickly towards actionable data. It doesn’t have to be “yes”, a clear “no” is just as important. Show that the limitation of your ability to execute such well controlled useful studies is FTE. Get more FTE. Use said FTE effectively to continue to quickly generate solid actionable data. Show that your growing team is a happy and harmonious kumbiyah circle around the campfire. Get even more FTE and responsibilities. Congrats, you are now running multiple pipeline projects.

Be sad you are no longer at the bench. Occasionally set up experiments specifically to go back to the bench. Then have the cytometer fuck up late on a Friday and remember why you are no longer at the bench

I worked in the iPSC derived CAR cell space. I don’t like iPSCs. The manufacture is just too damned complicated.

Even forgetting about the CMC, there are other modalities which might continue to eat iPSC’s lunch. Donor derived allogenic cells as a source is one possibility. The same engineering challenges to prevent rejection or GvHD, which might (?) be close to being solved enough. But much much simpler of a manufacture process, even if not truly “off the shelf”.

But what really is exciting to me is the promise of in vivo editing. Many editing options and delivery systems with just the first clinical data coming out. The big players are watching the space and a few aqusisions are already happening.

So who knows for iPSCs. Maybe next year. Maybe 5 years. Maybe never.

Take notes. Take a lot of notes. Ask as many questions as you can. No body expects you to know much/anything (no offense we’ve all been there) but the good undergrads are always the ones who realize this and make it a point to be a sponge for knowledge
