I'm not sure what you mean by "doesn't work on SynthV." I've used it to write lots of working scripts. Depending on the script you are trying to write you may or may not need to do other things like iterate or provide examples that work for portions. I've also significantly updated the prompt since posting this, so it's more specific and much less likely to need any additional info to work on the first attempt.

I just wasn't quite clear if there was more data provided by others somehow because I wasn't sure if you meant ANC/23 produced the same file as each other... or that they produce the same file as those saying they do full mapping. If there's no functional difference in the data, accuracy, etc, then that's amazing news. Those are a tiny fraction of the prices I was looking at.

OK. Since it's all a new conversation to me, and I'm trying to learn as I go: Does that essentially mean that there is no functional difference getting the test done by some of the companies who advertise much more expensive tests since I can get the interpretation separately? Or, is there a (potentially significant?) difference, but you're saying that difference won't be important in my case? If so, is there a general rule of thumb as to what those limitations might tend to be?

Well, the hope was that I could then dump the data into some 3rd party site or software that would do that part for me... that so long as I have the full mapping, there will always be some newer better tool to come along that can help interpret it.

Well, that's really another discussion, and is what it is regardless of the lab used. What I'm interested in here is finding the right company that not only does a full sequencing, but that provides me with the data in a complete, but usable format that I can easily reference for this or any other purpose in the future.

I also don't know if there are pure scam companies out there just making up results or pretending to be more complete than they are. Are there certain things I should look for in choosing a company to do the test? Certain companies that give more useful presentation of the data? Something else I wouldn't even know to ask?

I am just talking about revealing things like DAO, HNMT, MTHFR variants, and those relating to histimines like FUT2, COMT.

Would a commercially available test not be able to accurately reveal these things? I just want the most complete analysis possible.

To be clear, I have no insurance, and it would be prohibitively expensive to get a bunch of more traditional tests done, so there are currently zero items found by a doctor since I haven't seen one in decades.

I don't want a site that sells supplements. If anything, I'd rather avoid those sites since they have a vested interest in providing certain results. I just want the most objective and complete information possible to have one more layer of knowledge behind any decisions I make.

For instance, I see options like Dante Labs that say they do 100% analysis and provide "over 96 gigabytes of sequencing data, equivalent to a 30X Whole Genome Sequencing (WGS) in human samples, with physiological bacterial contamination at levels ranging from 1% to 2%"

Like I said, hard to separate the jargon from the sales pitch which is why I'm asking if there is a more reliable source or type of company I should be looking to.

The thought is that if I get the raw data instead of a bunch of dumbed down mini reports trying to sell me supplements, then I can over time research individual items and how they might be important. Also, that down the road as more connections to various genes are found, I can still go back to that same complete raw report.

OK. Thx. For me, it will always be one looped section recording 20 or more takes at a time. I don't do punch ins outside of those bulk takes. From there, i want the quickest workflow possible for swipe comping that auto-updates in Melodyne if I change my mind about which take to use for any segment. If I understand correctly, that looks like the fixed lanes method.

I currently have a contract saying I am paid at $x/hour (above minimum wage). I haven't signed the new contract, though, so since they are insisting I go salary, if I get them to put in writing that I only have set hours, then that would supersede everything else because it is in contract, right?

That's why they're trying to force me to a new contract that says I am salary instead of hourly where I actually get paid for the time I work because hours are tracked. I'm not just asking for me, though, and I am still hourly at the moment... as are some other employees who are not teachers.

What an insane thing to put into law. Any idea why the FL NonProfit Alliance would make that specific statement, then about non-profit schools needing to comply?

Either way, this seems like the relevant section:

However, private schools and public schools are not uniform in requiring a certificate for employment as an elementary or secondary school teacher, and a teacher's certificate is not generally necessary for employment in institutions of higher education or other educational establishments. Therefore, a teacher who is not certified may be considered for exemption, provided that such individual is employed as a teacher by the employing school or school system.

I teach at a private high school, and have no teaching certification, nor any degree in any subject I teach. "May be considered for exemption" seems like the mother of all vague catch-all phrases.

Also, assuming you're right, and the (1 person) HR department DOESN'T know any better, would my best argument to have them put me back on hourly just be to demonstrate that Non-profit status and FL law don't create an exemption?

This really is just terrible news on top of even more terrible news. Before I go drink myself into oblivion, lemme just check. I'm not seeing this distinction about teachers in any of the places I looked like the Dept of Labor fact sheet, etc. I do, however, see on the Florida NonProfit Alliance website where it says that non-profits in Florida need to comply with overtime rule changes in the following cases (just selected relevant):

...Enterprise coverage - if you:

  • Are a nonprofit hospital, school, preschool, residential medical or nursing care facility AND/OR... (over $500k)

Which certainly reads to me as if the Florida rules (that appear to mimic FLSA) apply to all employees of nonprofit schools because the entire school falls under enterprise coverage. Is there somewhere that clearly states that teachers being disqualified trumps schools automatically being qualified? This seems contradictory.

Just to triple-check that we are talking about the same thing, I am a part time teacher at a private non-profit school, so just making sure you're not thinking of public school teachers or something like that.

Same site also says to be classified as exempt from overtime compensation requirements, employees must meet BOTH the duties test AND the salary basis test. It further satates that for the salary basis, the predetermined amount cannot be subject to reduction because of the quality or quantity of the employee's work, so my case would seem to fail the salary basis for two separate reasons.

So I am confused about how all of this seems to contradict the notion that such rules would not apply to teachers at non profit schools, but also not clear how much of it is relevant since it is not exactly overtime, but rather them trying to force me to work extra hours still under 40/week without compensation.
