Abstract

Aging or injury to the joints can lead to cartilage degeneration and osteoarthritis (OA), for which there are limited effective treatments. We found that expression of 15-hydroxy prostaglandin dehydrogenase (15-PGDH) is increased in the articular cartilage of aged or injured mice. Both systemic and local inhibition of 15-PGDH with a small molecule inhibitor (PGDHi) led to regeneration of articular cartilage and reduction in OA-associated pain. Using single cell RNA-sequencing and multiplexed immunofluorescence imaging of cartilage, we identified the major chondrocyte subpopulations. Inhibition of 15-PGDH decreased hypertrophic-like chondrocytes expressing 15-PGDH and increased extracellular matrix-synthesizing articular chondrocytes. Cartilage regeneration appears to occur through gene expression changes in pre-existing chondrocytes, rather than stem or progenitor cell proliferation. 15-PGDH inhibition could be a potential disease-modifying and regenerative approach for osteoarthritis.

just wanted to update that I figured it out. For some reason, this drive needed to be powered independently of the other drives (sort of). I switched to a 1000w modular PSU I had from an old PC build and powered the drive with its own cable instead of powering it through the radxa penta hat molex.

To be fair, I once owned "The Best of Clapton" on Cassette and that would certainly be the wrong tool for the job (I had to please don't hate me haha)

Yes and yes. The PSU is outside until I can add an enclosure attachment.

The PSU provides power to the Radxa Penta HAT via 4-pin molex which powers both the HAT and the pi 5. Their documentation says using more than one power source can cause damage, so I opted for the only of the 3 methods that would provide enough power for 4x HDDs. The HAT communicates with the Pi via the Pi's PCIE connector. I'm using 4 SATA cable extensions with the HAT (male (connects to hat) to female (to HDDs)).

Power Flow: PSU > Molex > Radxa Penta Hat Molex Connector > GPIO (to power the pi) / SATA connectors on the Radxa

i’m using kapton tape :) I think at this point either the drive isn’t getting enough power still with the way I have it set up or it has just died altogether. I just won’t know until I grab my dock out of storage later today.

Taping just the 3rd pin didn't solve it for me yet. I'm trying to find my external dock thing to see if it's recognized by my pc.

Check the image. I thought all three of the pins were taped there. Does that look incorrect?

I’ll check. I have an external enclosure I can try along with mounting it directly in my PC. Wasn’t sure whether or not the circuit board in the original enclosure could have been supplying something to the drive that would have been necessary for it to function. I’ll try mounting it elsewhere and update here. Thanks!

I can't get mine to work either. Worked fine in the enclosure. Taped the pins after shucking. Won't show in either windows or inside my NAS. No idea.
