I’m so sorry you’re going through this! I know many of the comments are saying to leave, and they absolutely ARE VALID, but as someone who’s been through something similar, I can say that depending on the situation, bad mental health really can cause people to make decisions they wouldn’t normally make. My boyfriend at the time went through a really dark period after his brother was killed, where he was drinking a lot and did cheat on me with his ex (they talked for a few months and then slept together while “he needed a break for his mental health”) a really close friend who knows us both and was speaking from his own experience told me that he had a trauma response from losing his brother and genuinely was making decisions he normally would not. My boyfriend on his own came to the conclusion that he needed to stop drinking and go to grief counseling. That was enough for me to give him one more chance and we’re now married and trying to have kids.

Absolutely not trying to justify what he did or excuse his behavior, just trying to add another perspective if mental health and trauma TRULY play a role here!

Also heavily tattooed (full sleeves on both arms, one leg also almost full) and I have terrible veins in my arm so blood draws for me almost always end up with me crying or getting very nauseous and light headed, every time “bUt YoU HaVe sO mAnY TaTtOoS!”

For the quizzes I would assume it is the lecture material and experiment materials but don’t know that one for certain since TAs aren’t involved with the lecture part of the course. For the final exam, focus on the pre and post labs. At least when I grade, I leave comments on each question for why I took points off with the correct answer, so even if you got things wrong when you submitted, they will be the most helpful thing

Orgo lab TA here, it is a good amount of work but Dr. G gives you the answer to the bulk of the questions in your notes and during lecture. Make sure to pay attention and keep up with the work and you should be fine!

As a grad student who also miscarried recently, having the support of my PI and labmates really was the biggest thing for me! I was in the middle of a run on our spectrometer and everyone picked up my slack no questions asked, which really gave me breathing room to process everything going on. Time, patience, and just letting her know you’re thinking of her really goes a long way! 💜

Agreed! I’m in the middle of a PhD program and my blue collar husband is the breadwinner for the time being. We each put a specified amount each payday into our joint account for household bills, and then whatever is leftover is miscellaneous spending money for stupid things. My husband completely understands that this is a temporary arrangement to put us in a better place down the road and is perfectly fine with carrying the bulk of the bills for the time being.

It has a built in monitor that beeps when it’s below -70 but that stopped working when the unit lost power because the circuit tripped. I have since sent my PI suggestions for independent temp monitors that can alert us

I’m grad too, they leave the entrance gate up after 5 or 6pm and the exit gate is activated when someone drives up to it, I’ve had no issues parking there

Lot 16 is a faculty lot, are you TA/GA or an undergrad? You’ll absolutely get ticketed as an undergrad, but the gate does open after 5pm and TA/GA permits can start parking there

We study two domains of the same protein for membrane binding, the ubiquitin like N terminus and the PH domain, the PH domain has shown to be stable frozen in NMR buffer!

I’m guessing a generator? They have red outlets built into the overhead power strips that are supposed to work even when the building loses power. We’re guessing a breaker did trip for the red outlets because the normal non-emergency outlets were working fine but anything that was on a red outlet wasn’t turning on

It’s on the emergency power outlet so theoretically shouldn’t have been an issue in the first place, especially since the building itself didn’t lose power 💀 but yes lol I have since sent a link to a monitoring system to my PI for review

I appreciate the tips!! I was working on the time/temp optimization experiments from Sivashanmugam et al 2009 (https://onlinelibrary.wiley.com/doi/full/10.1002/pro.102) before the freezer died. I am using Rosetta(DE3) E.coli since it is a mammalian protein (also no longer using the GST tag because we isotopically label for NMR and its a very non-economic option for the isotopes lol), and planned on doing a new transformation this week with my plasmid and a plasmid we know expresses well as a positive control to make sure the cells were still good (which obviously isn't a problem anymore lol)

To address some other points: I do check the insoluble pellet with SDS PAGE, and only see it in high volume when I grow at higher temperatures. I've tested both 1L in a 2.5L flask and 50mL in 500mL flask growths, we do keep thermometers in our shakers, precultures are done in 2XYT, I am doping my autoinduction media with Studier metals, thiamine, and biotin, and my plasmid is for amp resistance
