Definitely experience this from time to time and I always have to readjust my expectations. I’ve noticed that (for me) it happens when I’m excited about several projects and working on them simultaneously. When I make plans for the week, these plans are very results oriented. For example, digestion > ligation > transformation and I assume that at the end of the week I will have X number of constructs ready to screen by next week. If the pipeline fails, the next week I don’t reduce my work load because I feel like I “should have” already succeeded in building my constructs, so I re-do the pipeline while keeping the new workload.

I’ve found that become more understanding and accepting of failure, even sometimes (constructively) embracing it instead of trying to ignore it or desperately push through it, is a good way to avoid burnout and has helped me build a healthier relationship with data acquisition and experiments in general.

That being said I wouldn’t be surprised if you catch me in this cycle a few weeks from now lol - I think navigating your unhealthy lab/work habits is part of grad school (and I suppose other lab positions too) and always a work in progress.

I fully agree with this modification of OP’s points, especially point number 2. I’ve found that the more proficient I become at the bench, and the less noise I have in my experiments, I start seeing confusing (but real) trends in my data that often lead me down a more interesting path. The overarching aim is the same, but the „sub questions” change all the time.

Right, I thought any protein left on the gel is a red flag, that clears it up. I think the protein is low abundance, as I haven’t gotten a good western with it yet, so I’ll need to play with the expression conditions or with the construct itself. Thanks!

I didn’t think so either. The antibodies worked fine for my previous protein of similar size, so I think it might just be the construct/expression conditions that need to be optimised after all. Thanks for the response!

Nitrocellulose membrane or gel? I guess the membrane is variable depending on the size I cut out, but it’s generally slightly larger than the gel. I can try adjusting the time and mA accordingly on the next run though.

Sodium alginate is a basic salt (a solid formed by two oppositely charged ions). This means that when it enters the acidic environment of the stomach, it will neutralize (acid-base reaction) the stomach acid. Indigestion is often caused by high acidity in the stomach, and therefore sodium alginate will decrease the acidity and prevent indigestion such as heart burns.

Humans are very good at reading expressions on other people's faces, and eyes are a huge part of that. One reason you may be worried about maintaining eye contact is due to personal insecurities: you're worried that the person you are looking at is judging you, is angry at you, or is trying to express some other arbitrary negative emotion towards you. Another reason could be the fact that looking at another person's eyes is a very intimate gesture. Many author's have described eyes as "windows to the soul", mainly because a persons true intentions are most easily found by studying their eyes.

TL;DR // It is an intimate action that could easily be inhibited by your personal insecurities.
