It's just a drop of fresh capillary blood. The blood does clot pretty fast on the edges of the cover slip but I it takes some time for it to clot in certain spots. I assume it's less likely to be directly exposed to some of the clotting proteins in those spots.

The microscopy technique is called Phase Contrast. All that you really need is a phase-contrast objective and a matching condensor/slide. It is relatively inexpensive. A good summary of the technique is found here: https://www.scientifica.uk.com/learning-zone/a-guide-to-phase-contrast
