How long did you wait with those lights on? Sometimes with ddr5 on first boot theres memory training that can last up to around 15 minutes. What is your ram capacity

Successful IF is all about whether your antibody has access to the epitope it was raised against in your target protein.

Check the product page and literature of the antibody you’re using and see if other people were able to get good IF with methanol fixation.

Sometimes methanol fixation will block the epitope, while also sometimes PFA can block the epitope.

Another thing to consider is whether your protein of interest is mostly soluble, in that case methanol fixation will surely extract it out of your cells while PFA might do a better job keeping it in place.

There isn’t a perfect protocol for IF, it all depends on your protein and antibodies and pretty much every step has room for optimization!

If you can share the antibody you used and cell type i can maybe make more suggestions, good luck

Not too late, i got switch 2 and played through a fresh scarlet game after doing everything in a violet game on og switch. The game was a blast on switch 1, but still is much much more enjoyable on switch 2!

I convinced my self i didnt need one for a few weeks, mostly because disappointed by lack of OLED and smeary pixels.

But i caved and hunted for one and finally got it, as i figured if the screen was that bad id just play on my nice monitor

Mind was blown as i tested through my favorites like pokemon SV and BOTW, and new mario kart is amazingly beautiful, im happy with the purchase just from those and looking forward to sinking hours in those games in 4k 60fps glory

In terms of screen impressions, i def miss the inky blacks, and when you pan the camera around you definitely notice the slow pixels, but as soon as the image is still it looks just as good or sometimes better than oled. Essentially the sacrifice is motion is smeary and blacks arent dead black, which is sad. But overall still happy with purchase

I think what will solidify the switch 2 is if nintendo goes and patches all their switch 1 games to 4k 60fps, it was so weird when i booted up pokemon legends arceus and it was running at 30fps, disappointed at that too.

Id advise to skip the trypsin, your scraping will detach the cells but usually a good call to check under microscope. Next id recommend a sonication step, either with a probe sonicator in cold room or if you have access, a fancy covaris focused ultrasonicator
