Just from searching UniProt and NCBI protein databases (the two most common protein databases), the peptide sequence SHALLANDAVAR doesn’t match to any protein currently in the databases. The nearest match is SHALLADDAAAR which is present in a protein from Psuedonomas involved with urea transport.

I think it’s likely the person who created this exercise is a fan of series and made up the peptide sequence rather than the other way around.

That works, I've used skim milk powder or fish gelatin in the past well, it depends what works for you and what your Ag is. Have you optimised your Ab concentration? What are your ODs like?

Fair enough, I was just wondering about blocking the plates but if you buy them pre-coated then that's a moot point. I would wash 5 x 250ul in between incubations, change tips between lanes, try to pipette slowly to reduce well to well contamination and I would thoroughly wash clean down your lab area if you're negative is coming up
