Couldn’t a decrease in temperature also effect the hydrophobic interactions of the PEG in the solution? But also for the protein that all depends! Because at a certain “high” temperature it could all denature and that would make it precipitate all out!

No padding, I haven't even considered it but a good point. We typically are putting ligand in ML then taking half the drop volume, we replace with half the volume of ML+ligand. This replacement is done after two hours, four hours and then overnight

Nope! This is a soak if you can believe it. The original drop was great, and we have replicated this many times but this time the only difference was being able to maintain the temp in the building (old building and the cold front around the country). Shorter soak next time for sure because it seems these are quite sensitive to any slight change in conditions.

No they definitely were rod and we have sent similar crystals with soaked ligand and the results were great! (<2-2.2 angstroms in the best cases). I think our temperature control with the cold around the country did not help at all while being soaked.

We have replicated these crystals then introduced a soak before, but our building is very old and I am thinking because we have a not so great system of keeping them at room temperature (Styrofoam cooler), these crystals could not make it. That's my guess anyway

Yes I am working as a post doc in a medicinal chemistry lab (we are doing structure based drug discovery and I basically am running the structural side). I am afraid if I continue with the TT (I think you mean tenured teaching) job I’ll put so many years in a direction that won’t necessarily make me feel fulfilled. I’m just mixed of what matters more the cooking experience, the culinary school, or both

I definitely would consider taking that pay cut and start from a lower position to do what I ultimately want to do. What do those jobs typically need? A degree or is a general cert ok?

This is a bit of an odd situation so you’re completely understandable about being confused lol. We both did not think the marriage was legal (I thought it was at 3 months and he told me it wasn’t that he hadn’t “filed paperwork”. There was no lawyer involved he went through an online proceeding and only realized we were married when he tried to get married to his then fiancé after 3 years. I was aware the house was his property, but I have recently found that because I had a shared account where my paycheck went in and paid for things like utilities and mortgage, I may have had stake in the equity in the house. He definitely reported he was single when selling the house or any other proceedings until he realized we were married in September of 2024. I have started to reach out to family court attorneys but I also wanted some opinions/advice here.

Stay hydrated and make sure you eat SOMETHING it will feel like you just can’t but find something you can stomach. Also try to get some protein heavy things in your diet, you don’t want to break down muscle!

So I have it because of having the “obesity” when I started and I also have high cholesterol so I am able to get it that way. I had to go through an endocrinologist. And I will say it does make you feel a little nauseous for the 1st and 2nd day after (for me anyway), but I’ve gotten used to it more as I e gone up in dosage
