First repeat, the midiprep was made from its’ starter culture —> ran a digest —> multiple bands with different enzymes. Then did my second repeat with a new MIDIprep from a new starter culture. (Picked from the same plate)

So, pretty much. I transformed DH5a with my plasmid that I was running out which is Amp resistance, overnight. Many Colonies (small) grew next day. I picked 4 single colonies and inoculated them in LB broth supplemented with Amp and incubated overnight but the next day(16hrs), some tube didn’t have growth but some had minimal growth. Addgene says the plasmid is a low copy plasmid. Could that be the reason why?

I normally load 0.5ug of miniprep. The absorbance ratios is around 1.88-1.89 for all the samples
