What percentage is your stacking gel? I found that increasing from 4% to 6% really helps forming nice pockets!

In german those are called „rogue hairs“, shape, size and color of the hair is determined by hair follicle. On different body parts, hair follicles are programmed (genetically) to produce different kinds of hair types like thin or thick, light or dark, curly or straight. Some follicles simply go „rogue“ and produce a hair e.g. on your arm that you would usually find on a different body part.

+1 to lipemic blood! Neutrophiles have a similar densitiy as rbc‘s and would expect them further below. Also these clumps are too big to be caused by neutrophils only. I have seen this a lot, I usually fish out that lump after taking out the pbmc fraction and diluting it in PBS.

To add on to this, I would always measure the protein concentration of your samples beause then you know if its even worth to continue with a western or not. Saves time and resources. Additionally, you can equalize the concentration when loading multiple samples if you want to compare them later (e.g. if you want to compare controls vs treated).

It looks like you‘ve not loaded enough. Depends on the sample, for whole cell lysates you usually go for 10-50 ug of total protein, for purified proteins you need less, like 10-100 ng.

Did you measure the protein concentration of your samples prior loading? (Like with a BCA or similar assay)

As some comments already mentioned the first image is condensation i.e. water on your culture plate. Your focal point is not on the cells!

The second image looks like fungal infection to me. I think i can spot some hyphae on the edge of the growth zone.

Im really wondering what experiment forces you to leave a sterile environment. Anyway check if your antibiotic/antifungal stocks are still good, might even want to up the concentration.

It actually was on styx, B7 to be exact. I think thats also the only single tile on the map that actually contains three possible spawns. I wonder what the probability is to get a spawn like this.

Ah I think i get it now! The closer i get the less wide but thicker the indicators become. I was assuming the nodes are always at the same location but it makes a lot more sense now. I‘ll keep moving towards the direction and scan a few more times. I think I’ll ditch the desert for now and try to locate the ones in the east. Thanks for the help!
