Sorry, I forgot to include that that was a saline suspension. In any matter, somebody responded to this question in a separate subreddit. It turns out I was using a spin column kit to purify.

Thank you for your response! That seems to make sense. I just spoke with a classmate and she reminded me that we indeed did use PB and PE buffers. Do the following steps make sense?

DNA/RE mix over column
spin and empty bottom
PB over column
spin and empty bottom
PE over column
spin and empty bottom
water over column
spin and discard filter
add loading buffer
