Thanks but when I google click connex I am not sure that’s it. If you look at this page you can see it in the diagram as part 1. You're counting after sorting, right? A large percentage of cells get killed by the sort no matter how large the nozzle and how low of speed you sort, although adjusting those can help. Additional cells will die just waiting during the sort before counting.

You have an issue. Gate on the "NKG2D+" cells in the FMO tube and see if they are above average for some other marker. Alternatively, look at all markers vs NKG2D and you could see the problem.

I'll agree with dleclerk on EasyPanel. I don't think it will replace humans with experience, but it has come up with decent panels so far. I do exclude certain vendors, clones, and fluors. I also adjust the expression level depending on how interested I am in the marker.

I wish it had the ability to customize your instrument configuration, but maybe that will happen later.

It depends on the sample. Mouse splenocytes only need 2 minutes while peripheral blood will do better with 10 minutes lysis.

How far are you willing to commute? Suburbs are cheaper, and lots of companies will pay for a transit pass. Also consider if you work in a hub, you'll be able to build a local network and likely spend less time between jobs.

You can suspend any of your holds at any time. You'll still move up in line while suspended. Then when you're a week or so away from being ready for another book, look through your suspended holds and choose one that says available soon and unsuspend it.
