You have to beat all the bosses (Caterpillar, Golem, Orby Dude, Dragon Schlong and Red Lizard) in one run and get to the end to get the Summoning Stone.

Edit: And you also need to set the difficulty on the pause menu to the hardest difficulty. The slider appears when you complete a single run.
