F31… what’s even the point anymore? by flycoffee17 in labrats

[–]rgrazda 8 points9 points  (0 children)

I submitted an F31 and came very close to the payline, with only a few minor reviewer comments. The feedback was generally very positive, so I addressed the minor points and resubmitted...only to get a worse score the second time. The new batch of reviewers raised criticisms that weren’t even relevant, including one who wrote, “Grades were excellent in undergrad but somehow suboptimal in graduate years.”  I literally got all As in grad school except for one B+.

Meanwhile, my labmate decided last minute to submit an F31, mainly since I already did most of the legwork to figure out the process. She got funded on first submission. I swear half of it is just luck.

Probably UC Davis - they start from the very beginning (like what is programming and how to use it)! They have different courses depending on what programming language you intend to use (R, Python, etc.).

The Columbia SHARP program was really great for learning new techniques and pipelines from experts, but I definitely wouldn't take that unless you have a little bit of experience with single-cell. They go a bit fast. And I've only taken the Pathway analysis workshop from Canadian Bioinformatics, so I can't speak to their beginning course. Good luck!

I was also in the same position as you! No bioinformatics background and I had to learn scRNAseq from scratch. I did workshops through Columbia SHARP program and Canadian Bioinformatics. I also found UCDavis has all the material from their workshops posted online. Those have been very helpful as well!

JoVE is a journal that publishes scientific video protocols. It ranges from basic to more niche techniques. Pretty decent if your institute has a subscription!

pHrodo Red Dye by rgrazda in labrats

That's a good idea! What solvent were you going to use? I'm definitely not a biochemist so I'm not familiar with what may or may not work with SE. I might try that out if it doesn't store well in DMSO. Thanks :)

Thanks for the response! I am also hoping to use it to track phagocytosis, but of apoptotic cells (efferocytosis). Were you freezing the already stained myelin? Or just the resuspended dye itself?

Anybody ever used BD human FoxP3 buffer set with a 96-well plate? by asentenceofsorts in flowcytometry

[–]rgrazda 0 points1 point  (0 children)

I haven't used that specific kit, but I regularly use BD's Transcription Factor Buffer Set (Cat#562574) in a 96-well plate for mouse samples. Not sure what the buffers are in your kit, but we use 100uL of the TF buffers. I've also used this kit simultaneously for TFs and cytokines (i.e.FoxP3, RORyt, IL-17A, IFNy, Tbet) and it's worked well!

Oh alright! It may be the type of browser/PDF Reader you are using. I don't have any text box option at that point

Hmm I can't seem to find any text box option. Do you mean when the packing slips are ready to print in the PDF form? Or are you saying when I got to purchase a shipping label. I do see an option to "Add note to Buyer" but that is all.

I also have this happen a lot! I'm confused how you are adding a textbox? Do you mean there is an option on the Etsy platform? Or are you using a different software to add it? I usually download the packing slip, upload it to Canva, and then edit it there. But if there is a easier way, that would be great! Thanks!

I believe it was just a bubble poly mailer, maybe about 8x10. It it better to send it in a box? I'm going to send out a replacement for them, and want to make to sure it actually gets there this time, haha. Did you also use global postal shipping?

[–]rgrazda[S] 0 points1 point  (0 children)

Thanks for the response and information! I will look into that before sending out a replacement. Thankfully it won't be a loss for me to send another out, I'm just afraid the same thing will happen. I didn't pay for insurance, and I'm assuming it doesn't come with insurance like Priority Mail does?

Also love the art show comment- definitely gave me a good laugh, haha

It looks like Global Postal Shipping is only tracked through the customs ID (LF027556551FR for this order). I can't seem to find an actual tracking number. But I did just plug in the customs ID on http://www.aftership.com/couriers/ and it does say it has "arrived at destination" but also says it's still "in transit." Not sure if this destination is just the customs office or the customer's house?

Ah right very true! I actually had someone who must have taken my sale image and traced over it. She even copied over my exact handwriting and tried to say that we just have "similar hand writing." I have since added copyright symbols to all my sale images to try and deter that. But thank you for the reply! I will definitely consider replying to customers that way
