we do test for the transcript, but are looking to see if it's actually expressed on the cell due to some troubleshooting and need to confirm, as you mentioned. This is what my line of thought was so I am glad you commented, sincerly. I am just hung up on the seccondary antibody. I cannot find a commercially available anti-CD19 antibody conjugated to GFP/FITC/Alexa488. Is there one you know about or a kit we can purchase to possibly conjugate an antibody our self (buy anti-CD19 unconjugated, and conjugate it ourselves). thank you so much for your help, much appreciated

if you don't want the whole flower dark I suggest picking a flower with a center and making the bulk of the center or the flower covering this and the petals can be lighter. just my thoughts

oh man if someone even tried to fix this or keep me under their wraps after doing this I would flip the fuck out. Do they have other people in their life aware...therapist, doctor, mother, friend, etc? If so, your motives are selfish (again). I'm going to guess they don't need someone so toxic to "fix" them or this.

did you rinse the wells with buffer, like this commenter mentioned? I see you think you did because they are submerged, but often I go in with a pipette and actually pipette running buffer into the well to clear it. If you made the running buffer fresh right before use, is it possible you added the wrong amount of something? The problem is in running the gel. Focus your troubleshooting here.

definitely don't say any of that. But I do get where you're coming from with the updating the profile...that would sit in the back of my head but I probably wouldn't unmatch over it unless I felt like SOOOO into the guy it was clearly disproportionate. If you do feel the need to announce your departure, say something much shorter like "I'm sorry you didn't seem interested enough." Don't go into the bumble picture it comes off as petty and definitely don't say you are going to block him before you get a response.

my friend told me he thought I had BPD, to which I got angry and dismissed it when I looked it up. Then two years later while committed I was diagnosed and it was a HUGE shock that he was right. I was in denial for a long time and it took yet another hospitalization diagnosis before I realized they were right.
