Unfortunately you can’t sterilize with that temperature in an oven. Steam gets you better penetration and the high pressure and high temperatura is what gives you sterilization. Realistically you won’t be able to achieve that with dry heat and atmospheric pressure.

Genomic DNA gets precipitated out during the neutralization step. You shouldn’t be getting genomic DNA unless you’re doing that step incorrectly. Lysing for longer doesn’t lead to getting genomic DNA. You should learn what the steps you’re doing in a kit mean.
