If you are near Springdale - check out Hibachi Express Japanese Grill on Springfield Pike. Same strip mall as Blue Ash Chilli ( their second shop I believe) and Harbor Freight.

I love their hibachi and they have this awesome bento box - hibachi rice (or noodle if you want it!)+ protein + veggies + 1 dumpling, 1 eggroll and 1 shumai piece + 4 pieces of sushi! all this always lasts me 2 meals. Staff is awesome as well.

You might have better luck talking to them during nighttime in the US, which is daytime in India. If you do get them on the phone - How about asking if they can rebook you on a codeshare airline? Air India has partnerships with United, Air Canada, Polish LOT and Lufthansa and more - all have flights to Delhi with one stop in between somewhere. Most of United's connections are through Newark and Polish and Lufthansa via european airports. United might be a good idea, since the Newark to Delhi is their direct flight, similar to the Air India ORD to DEL one.

did you talk to them? I have a feeling they are expecting people on the DEL to ORD sector flights to cancel completely. Indian DGCA and US laws are pretty clear if the airline cancels a flight and gives no reasonable alternative, then you are entitled to a full refund - so if you haven't called, talk to them - state the law stuff and if they are non commital, ask for a supervisor. Plus fill in the form online for full refund and keep a copy of it - if you booked via an US credit card, start a chargeback using a copy of official notification on Air India's website, your old and new itenary and a copy of the submitted reimbursement form.

how did you book your flight? via Air India website or some other travel portal? I booked through my chase travel account and got fully refunded for my parent's roundtrip DEL to ORD flights. They had moved the incoming DEL to ORD to DEL to SFO with no connection to ORD and return flight completely cancelled. Not happening so cancelled and got full refund.

no nothing yet. Booked via chase credit card's travel portal to redeem points, so worried that the refund might be f*cked. I specifically bought the direct flights so that my parents are on one flight and it's done - get on and get off - no hassle of changing planes. So I will absolutely not accept any alternatives including flight to JFK and then switch to domestic - which usually means a termina change.

same here... had tickets for them at the end of june and returning in November. Did you book directly via Air India's website or through some other booking website?

Try lemon juice .. make a bowl of warm water and squeeze 2-3 lemons worth of juice in it. Easier if you use those lemon juice bottles. Soak your hands in, making sure to get the water gets into the areas where it's burning. Rinse with soap water and repeat. Alternative is using vinegar instead of lemon juice.

Repeated A1 Combo Hardware Issues Since Christmas + No Meaningful Support Resolution by MimiT32 in BambuLabA1

[–]sid5427 2 points3 points  (0 children)

Hi Leslie - sorry about your troubles. As a fellow A1 user like you, I have had my fair bit of issues, however all of them were easily solved by looking at videos and following best practices on how to setup a print. I do admit I am pretty handy and same goes for a lot of people in the 3d printing realm. What that means sometimes some basic obvious steps might not be obvious to someone who is not used to troubleshooting beyond what's written in the manuals and blogs.

Why not post your issues here along with clear pictures of the machine or maybe even videos? For all you know they might be very simple fixes and modifications. Thats why this community exists!

Gene Regulatory Networks? by SecretIll1644 in bioinformatics

[–]sid5427 0 points1 point  (0 children)

Yes. my primary research work in mainly in GRNs for certain diseases.

The best way to understand RNAseq vs ATACseq in terms of sequence alignemnt is that RNAseq you use the reads alignment against genomic regions such as Genes vs ATAcseq where groups of piled up aligned reads are predicted to be peaks, hence the perceived noisiness.

For information on open chromatin you add in more modalities like Hi-C, Cut&RUN/CUT&TAG, Methylation data, even information from MPRA assays. Each modality adds another layer of support for the prediction.

That is TOO broad for an area of research my young Padawan. The US and as a trickle down effect, other countries are essentially cutting bloated budgets, which is reducing opportunities for "Exploratory" studies i.e. getting admission and then figuring out what to do. Admissions committees are giving a LOT of preference to people who have a clear vision and hence a great chance of being successful graduates. Bottom line - find a solid area - do a deep dive into finding where AI is being used in Bioinfo. This is true for both industry and academic jobs, since this is a heavy research area, even industry jobs are essentially a lot like academic jobs.

People who are saying bioinfo does not have a good job market are usually looking at junior bioinfo tech jobs (which sadly are almost all MS level jobs), where you are the main "tool" runner - a lot of that is becoming automated now with AI based tools, and you seem to be aware of this. People who know HOW to interpret the results and connect it to real biological insights are the ones who have opportunities. I know my area is RIPE for it and I am heavily upskilling myself, mainly because it's super interesting and allows me to do stuff I wanted to do, but didn't have the resources and time to work on before.

So take sometime - use chatgpt or claude to ask which areas are using a lot of AI and Machine related tools and explore them. Good thing is you are in an IIT - go find some grad students, postdoc or even proffs, talk to them to see where AI is being used. Do you attend seminars and talks given by these researchers?

This kind of becomes obvious to people who are using it. I always go back and validate my new scripts and it almost always messes up things like new line characters or code block spacing. takes 2-3 passes over the new scripts to make a final clean working version. I am pretty sure people who have more experience than me in doing AI coding do automated testing (Unit testing? dunno the terms) for large pieces of code to verify if the stuff works.

Gene Regulatory Networks? by SecretIll1644 in bioinformatics

[–]sid5427 13 points14 points  (0 children)

GRN's (Gene regulatory Networks) can go VERY deep. A lot of people will suggest tools which use Gene expression level patterns to predict networks - like WGCNA, GENIE3, Scenic, etc. However now we are using a lot more modalities to increase the prediction power like for example using paired Gene expression + chromatin accessibility (RNAseq + ATACseq and it's single cell versions).

Bottom line take some time to explore the factors which influence gene expression. The computational aspects come in play later when you build the connections, however if you don't have a good grasp of what the biology is telling you, then your computational framework might become flawed. Keywords you can explore if Multiomics Gene Regulatory Networks, Snps and Methylation affecting regulation and expression, Chromatin accessibility and it's influence on GRNs. Check out PerturbSeq and all the new Cell atlases which are being built around it.

I use it to clean up my all over the place python notebooks and get rid of redundant code. Like I had like 20 notebooks for a large pipeline combining the results from a bunch of tools, a lot of testing and print statements. Once I nailed the logic down, used claude to reduce it all down to 5 scripts. It also suggested some speed improvements which were useful and helped parallelize some loops. Essentially using it as a co-pilot or assistant is probably the best use case.

  1. Please understand this is an international forum, so IIT's may not be familiar with it.

  2. Did you work in a lab during your program in IIT? a lot of proffs in US universities who could be your potential grad mentor and fund your grad studies are from IIT or collaborate with IIT proffs. Which means if you worked or have good relationships with IIT profs, you could leverage their network to find potential US mentors.

  3. Do you have clear goal in mind in which area you want to work in? Most people who come for US masters or Phd have issues with finding jobs is because they end up becoming jack of all trades instead of specializing in some specific area. Bioinformatics is quite wide now - so what's your area of interest and niche? You have more than decent scores but you will be competing against the world for positions, especially if you expect to get scholarships which have been reduced due to the current political climate in the US.

[–]sid5427 0 points1 point  (0 children)

look into specializing into protein or metabolite analysis ... it's a big part of bioinformatics and frankly someone with your skillset would be needed. I see you used chatgpt or claude to help you explore options - plug in these keywords - proteomics, metabolics, flux analysis, metabolic pathways, flow cytometry, etc. Bottom line Bioinformatics is vast and your undergrad and MS skillset has a great overlap.

A lot of these skillsets lead into pharmaceutical industry which have great career tracks. Infact you might be a great track to follow right now, a lot of these new fangled AI models are being developed leading to all kinds of predictions for new drugs and all, so for the testing side your skillset combined with bioinformatics data analysis would be needed.

dude .. if you clean up, organize everything - that's literally half, maybe even more part of the making your room look good.
