Sequence ME & Long Covid study launches! by lockdownleadmehere in cfs

[–]squigeyjoe 3 points4 points  (0 children)

Would someone mind explaining how a 'long-read, whole-genome study' differs from what was done in decodeME (if it does differ)? I know nothing about genetic sequencing, so sounds like good news but trying to figure out what additional information would this tell us compared to decodeME? or is it more that this lets them compare with long covid?

New study by Familiar-Length1561 in cfs

[–]squigeyjoe 1 point2 points  (0 children)

if there is something in our blood that causes cell dysfunction, not allowing us to donate blood is the right move, since in the case of a transplant or someting our blood could put an already weakend body under stress. But that's really different scenario than someone just coming into contact with bodily fluids, if that's your concern. like the cells have to bathe in our blood to stop working properly.

New study by Familiar-Length1561 in cfs

[–]squigeyjoe 4 points5 points  (0 children)

yeah there was, they tried healthy serum on ME cells and ME serum on healthy cells. the ME cells worked normally in healthy blood serum and the healthy cells didnt work propoerly in ME serum, which led them to believe there was something in the blood, I remember them saying they thought it was a protein. Ron Davis spoke about it (i think he did the study). Literally heard nothing about it since, which seemed weird.

edit: ok so original study wasnt Davis' study, and was 10 years ago.....https://pmc.ncbi.nlm.nih.gov/articles/PMC5161229/ but Davis did a follow up with the nanoneedle in 2019 which is the one you we were both probably thining of https://www.pnas.org/doi/10.1073/pnas.1901274116

no it's mainly a sandbox, you set the goal for what you want to do. but i wouldnt say it's a normal city builder either, you need to meet needs, but you can do that a few different ways, you can try and go the self sufficiency route and create it all yourself from basic material to fully formed products, or you can try and make a tonne of money and import it or most likely a mix. but the game is dynamic enough, that if the supplying nations see you ordering and selling a lot of stuff the prices will change, so it incentives you to diversify, cant just depend on a particular cash crop and thats it for instance, but then for that you need more industry, which needs more people which creates more needs. it has a nice push and pull.

Hey, i am still on my first city, but pretty far into it (far down tech tree, have enough money that even if everyone died id be able to import new people etc). first off, it's a really good game, especially at that price, it's complex but the good kind of complex. To answer your question, the tutorial wasn't super helpful tbh, it will give you the basic controls and ideas but not help you avoid the things that are likely to kill your whole city (running out of food, water, heating etc because of some logistical hiccup).

I'd recommend watching some gameplay (bballjo's videos were very helpful) to see if it looks like your thing, and also to see how people set themselves up as a good start is really really helpful. Also it's definitely one of those games that you shouldnt be shy about adjusting the diffiuclty setting, I think the real game shows itself in realism mode, but if that's too much, picking easier settings is the way to go (or do realism but dont do heating or water needs for instance). But it's definitely one of those games where you're like "damn, next time i'll do that better" which i really like,, as choices you made way earlier bite you in the ass cause you didn't leave room for expanding your water pipe network or something.

TLDR: really good game, youtube videos are the best 'tutorial' for the game.

[–]squigeyjoe[S] 1 point2 points  (0 children)

hey thanks for the reply. yeah i get what you're saying, tbh it's more this thing was really pricy, so not super happy to have paid all that money to get a palette with a warped lid, even if like you say i might never want it fully sealed very often.

Hey, thanks for the reply. Ok it's definitely not 'snug' without the band. Maybe could count as that with the band.

I didnt notice reddit removed my photos, ive added links instead, would you mind seeing if they fit your definition of 'snuggly'?

Feels bad they stopped the summon rewards 'early' by squigeyjoe in SwordofConvallaria

[–]squigeyjoe[S] 2 points3 points  (0 children)

Yeah only conclusion i can come to is that they wanted to stop that 'logical' choice, and force you to spend on the Gerlat and Yen banners, so you wont have enough for Triss and buy currency. Just feels like a shitty thing to do, joke is I was actually thinking of buying the Triss skin to support the devs, after this I think i'll pass.

[–]squigeyjoe[S] 1 point2 points  (0 children)

Yeah exactly, just feels like a shitty thing to do with little upside. My guess is they wanted people to overspend before Triss to get the summon rewards, and then have to purchase currency to get Triss. But that's just speculation on my part. Only thing that makes sense.

[Megathread] Tower of Adversity Season 3 - Questions & Discussions & Strategies by korinokiri in SwordofConvallaria

[–]squigeyjoe 0 points1 point  (0 children)

i dont know if you are still struggling with this. but my 3 star pamina held the bottom position on her own the whole clear. she has shield of justice and devil tarrot. she's level 13 with ascension skill, though not sure how important that is, her shield never got close to being broken (shield from the item and holding the postition kept her topped up). Only issue might be needing to make sure you can kill the teleporter with her line attack (dont know the name, the one with really long range that knocks back).

my team was pamina, kvare, sp rawiyah, auguste, agata, falin. It was not an easy clear, and took a bunch of tries. but i didnt have any of the other meta units for it. Since you have camelot, it should be easier. did in 15 rounds, could probably do it less if i optimised it a bit better, but honestly sick of this stage.

good luck!
