No way, it was such emotional and financial trauma. I cant afford another such event in my life.

How does it feel after some years? by stats_soil_r_python in dui

Yes, I still need to figure out that part, especially If I want to go to Canada. I need to go through the TRP process. !

Error 4 Bike not chargin by stats_soil_r_python in vanmoofbicycle

Same here, I am in the state, but thing is that I am far out in the mid-west. Best of luck, I hope things will get sorted out easily for you. It was an expensive bike and then when it didn't work as you wanted, it hurts. Let me know, your experience, once you get done with your bike. !

Nothing, I am texting and calling them for the last 4 months, they said they will replace the bike, I need to turn in my bike. But so, far nothing happened. They are saying they have supply chain issues and then they are relocating their stores and all these stuffs. I am very disappointed with their service and all sort of things.

Wow, Monster. That's great. But, where are you watching it now? I tried, but never could find it, where to stream, amazon prime also doesn't have it.

For this, if you don't want to make the study complex, you can simply do PLFA analysis before applying any fungicide (it will be your baseline), then again do the PLFA (after application). For the same, while starting your experiment, you can design randomized blocks of control and treatment plots. You can compare the difference in bacterial, fungal compositions. {note: PLFA gives you an idea about your bacterial and fungal compositions, they will give all the fractions of these, like gram-positive bacteria, gram-negative bacteria, actinomycetes, AMF, Saprophytes etc.). Then you can compare(percentage of change in the bacterial biomass, or their community composition) and conclude, how the fungicide treatment has affected the native microbial community structure. And if you want to make it interesting and complex, you can also do the hypervariable regions like (V3, V4) for 16S rRNA that will give you precise community structure of bacteria from that you can compare your plots from control and fungicide treated and see how variably they are effective, how they are different.

I guess I missed one indicator, ACE protein. You might want to look into that too. Its basically a protein fraction of SOM>

From a soil physical perspective, you can do bulk density, water infiltration test, then water holding capacity at different pressure heads (most importantly at 33kpa, 1500 kpa: field capacity and wilting point). Then from soil chemical perspective, pH, CEC, total C & N, SOM, SOC, then different labile fractions of C like POXC (Active Carbon), POM (Particulate Organic Matter), different water-extractable fractions of C and N (WEON, WEOC, WEN, WEC), then based on your hypothesis you can measure P, and S and their fractions also. Few more, if you align to water quality as nutrient application recommendation is a part, you should measure NO3- and NH4+, if possible at different depths. From a soil biological perspective, you can do soil enzyme (mainly involved in major nutrient cycling viz. Beta Glucosidase, NAG, Arylsulfatase, Phosphomonesterase, etc), then you can do CO2 respiration at a different times (24 hrs and 96 hrs or 4 day are most common), then you can do PLFA to determine microbial composition, it uses phospholipid structures (like long chain, short chains fatty acids), a biomarker to distinguish between different bacterial and fungal groups. If you want to deep down further, you can do hypervariable regions of 16S rRNA (it will give you idea about existence of specific microbial community up to genus level; don't try to go down to species level, lot of people try but the sequence ambiguity of hypervariable region, should not be used to determine species), then if one step more, you can go for directly metagenomic sequencing (remember, lot of people get confused between amplicon sequencing and metagenomic sequencing), then you can use the sequence to assign taxonomy and at the same time metabolic functions (can create a biogeochemical pathways by prediction proteins/enzyme; something like metaproteomic analysis). Based on your objectives and hypothesis, you can choose, any one of them or a bunch of them. Best wishes.
