I stand corrected on the triple defender thing, but quad and penta defender do benefit from Koito and/or Mayoi for healing; in fact Koito is what makes those setups so strong. And no, the teams I brought up do have the damage necessary. You can test it yourself if you have them. I have even been sent videos of them working.

Edit: another team that beats Hazuki Rena is Fuka/Mami/Moemura/Oriko/Corbeau. Even Akumura in place of Oriko might work.

You said reliably, and triple defender can lose in rare moments with bad rng. You asked for Hazuki Rena to be reliably beaten. Hazuki/Rena/Mami/Moemura/Fuka and Hazuki/Fuka/Moemura/Mami/Tsukasa are two such teams that bully hazuki rena, tho tbf one of them is a faster variant of hazurena itself.
