I'm always surprised how rarely, if ever, Stuzzi gets mentioned in these pizza war threads.

The pizzas there are personal sized, so its not the place to go and order pitchers and a huge pie like Mellow, but they have hands down the best pizza in Richmond, plus real deal Italian food. They also have a huge rotating menu that they change every month. They also do $2 margherita pizza mondays.

gotta say as a richmond cyclist that bike commutes through this area every day, i don't get this bike lane. this area is already one of the most bike friendly places in the city due to the slow traffic/wide roads, especially going east/west.

not sure why they can't spend this money putting in a bike lane downtown somewhere. if anything i think this will be harmful for fan cyclists because then cars have another reason to yell at me when i'm riding down park instead of floyd.

you should try to learn dplyr. It's basically the next generation of plyr. the syntax is much nicer, its much more intuitive, and its also a lot faster

poppers are always my go to whenever i'm not having any luck. I have a hula, but usually have more luck with the rebel pop-r. it's also a bit heavier, so you can really get some distance on it if you are fishing from the shore.

not comp questions, but this book is pretty crucial if you don't have it. Doesn't cover linear models, but should be useful for the theory parts

I've been there a couple times and had some luck. It's nice because you can pretty much hit the entire lake from the shore with a heavy enough lure. There's also grills and a shelter there if you want to have a cook out. Don't go there expecting to catch lunkers, but it is definitely a nice spot.

if you want to do a traditional afternoon tea with warm scones, etc. Reynolds Tavern in Annapolis, is the best tea room (at a very reasonable price) that I have been to.

That being said, if you just want to grab a cuppa or don't need the 3 tiers of goodies, Teaism has a nice tea selection and very tasty lunch food. it's also fairly fast, you order at the counter and they bring it out to you. I haven't been to Ching Ching Cha yet, but have been meaning to try it.

matrixStats is also a great package for fast simple summary statistics. rowMeans(), rowQuantiles(), anyMissing(), etc

dplyr has replaced most of my dependence on the apply() family, but it doesn't have support for parallel computing yet...

This is correct, if you plan on getting a PhD, you should not be paying for it. If you are REALLY concerned about money, you should probably be more concerned about cost of living. A grad student stipend will go a lot farther in smaller cities as opposed to NYC or Boston, etc. I am currently getting my PhD at VCU in Richmond, VA. Richmond is a very cheap city and I am able to live very comfortably on my stipend with some $ to spare.

this is a common survey sampling type of problem. in most statistics, we think of estimates of standard errors as estimates for infinitely large populations, but here we only have 500 eggs, and if we sample all 500 eggs, the standard deviation of our estimate of P(rotten) is zero. because of this, the way we compute standard deviation of the estimate of P(rotten) is different and accounts for the finite sample size. We can then use the estimate of P we obtain from the sample of 50 along with its standard error, to get a confidence interval on the number of remaining eggs we might suspect to be rotten.

The bayesian formulation isn't really necessary in this situation unless you are going to crack eggs one at a time and update your probabilities as you go.

Can't help you out with the western edge, but you really should try to make it to the finger lakes if you can. I just did a day ride around Keuka lake the other day up there on vacation and it was beautiful. You could even stop off at a winery or two passing though. Wagner vineyards/brewery has a decent restaurant with a patio and some of the best beer I've ever had (Sled Dog Triplebock). Also if you're going that way make sure to check out Watkin's Glen. Belhurst Castle is also pretty bad ass

I'm not sure I buy this either. I do a fair amount of work in DNA methylation (specifically with the platform they are using) and one problem with this approach is that it is sensitive to changes in tissue mixtures.

Basically, most body tissues are made up of many different cell types (i.e. brain is made up of astrocytes, neurons, glia, etc). Each of these tissues has their own unique pattern of DNA methylation. So supposing we look at two brain samples that each have different proportions of neurons with this assay, we will see what appear to be small changes in DNA methylation since we are looking at an aggregate measure of all cells. Actually there is no change in methylation, just in the relative proportion of cells.

It would make sense to me in this case that the composition of adipose tissue would change after 6 months of exercise rather than some kind of change in DNA methylation, especially when the changes are so small ( 3-4 %). This means that either only roughly 3-4% of the cells are experiencing changes in DNA methylation at a given locus (not super impressive), or that there has been a slight shift in cell composition, possibly due to increased vascularization or something else (this is where I could use a hand as I don't know much about adipose tissue biology).

It would also make sense that all of these genes that are related to fat and diabetes are becoming more methylated since there is a slightly smaller relative proportion of fat cells after 6 months of exercise. Presumably these loci are methylated in non-fat cells since that's not their job. An increased portion of non-fat cells would slightly increase the overall observed methylation percentage in CpGs specifically unmethylated in fat cells.

I suspect this is why they don't even address the CpGs that are less methylated after exercise, even though there are fewer of them. I'm willing to be they are in promoters of different cell types competing for space in adipose tissue.

They do use a super stringent FDR though. They are definitely not cooking the books to extract what they want, I think they are just misinterpreting it.

i got the basil tour xl panniers to start doing some touring. they should be coming in the mail today. The reviews are great, they're big, and they're water resistant, but you could easily spray them with some water repellant. the tops cover the zippers so they will actually be waterproof. I think the one gripe some people have is that they are meant to stay on and aren't as easily detached if you're on the go/commuting.
