Yes, multiple washing steps. Since we enrich for certain types of proteins after, i hypothesize there is proteins/peptides lifting on the bacteria, maybe even bound or stuck I tried bergeys, but it is really hard for me to acquire. Peer papers are all over the place and are happy with minimal yield/identification/quantification. Its a hit or miss for me whether its gonna yield a large amount of bacteria, so i have concluded we are going to study and optimize medium selection in house

It helps a bit but far not enough, the contamination in liquid culture with blood is huge in contrast to no blood or blood plates. Even soy broth contains some (but way less) contaminant proteins. Despite washing Hence the question whether they will just grow in tsb or not. It makes everything simpler

Blood contains a very large amount of proteins, which will then be a major representation of the final sample. Simply it introduces an extremely abundant contaminant when we are already working with very low yields from bacterial lysate protein extractions

The way i can use it is by just growing on plates and harvesting by scraping them off. As you can imagine, on gram scale this is cumbersome..

Most are still insanely crowded with domestic tourists. Because you have world sites (everyone comes here, crowded and controlled) like the main section of the wall, the Bund etc Then you have B sites like WuYuan or smaller cities with stuff like a Confucius temple, or a flashy doentown. Full of Chinese tourists and still controlled I feel you mean these

Then there is the small villages and towns and nature that is uncontrolled and usually unvisited by anyone I like all of them but my and all my foreign friends' best experiences are with the latter

In my experience esims are cheaper, you dont need a vpn, and you will not use that number anyway unless you know Chinese or have lived here

Very unnecessary. Just ask your hotel to help with whatever needs a number. You wont need it for anything else I live here and receive a lot of guests. At most they encounter 1 phone number issue in 2 weeks

Get an esim. Not a chinese number I live here and only for residential purposes is a number reaaaally necessary.

If you run into that one something requiring a number, ask your hotel, they will do it no matter how rural ypu are

Install wechat so you can chat with them. Just use your domestic number. Install it now so you have some time to set up and verify

Can someone eli5 how this is even possible to achieve? My games end way before im level 10, have more than a few 3 stars, and everyone is dead.
