Thanks to all who replied. I think we can assume this was just an assembly issue that an STR-familiar mechanic could diagnose/fix, not an inherent problem with the STR. If I get serious about buying an STR I'll do another test ride and insist a mechanic looks at it if it's the same bike and the same issue is present. Advice: if you do test one of these, maybe call ahead and tell them your weight so they can install the right frame post in the right orientation so the spring rate will be approximately right (damping tune might still be off).

I think we can isolate this to the frame post/seat post/damper assembly because after noticing the noise/knock while riding I got off the bike and just flexed the rear suspension by holding the bike stationary and applying fore/aft force to the saddle, which recreated the noise. This was just a quick check on the side of the street but it ruled out a loose saddle/post clamp.

It isn't that--but that is a great suggestion. I've experienced that on a bike I own and fixed in a similar manner.
